The combination effect of Prominin1 (CD133) suppression and Oxaliplatin treatment in colorectal cancer therapy
This study was designed to check the combined effect of CD133 siRNA and Oxaliplatin on proliferation, migration, apoptosis, and stemness properties of CRC cells in the HT-29 cell line. MTT assay was performed to define the combined effect of CD133 siRNA and Oxaliplatin on the viability of HT-29 cells, and it showed that the combination of CD133 siRNA and Oxaliplatin could reduce the IC50 of this drug from 32.85 to 19.75 nmol. In order to figure out the effect of this combination therapy on CD133 expression at the gene and protein level, qRT-PCR and western blot were exploited, respectively. The results demonstrated that th...

CDK-associated Cullin 1 promotes Cell Proliferation and inhibits Cell Apoptosis in Human Glioblastoma
CONCLUSION: CACUL1 can promote cell proliferation and suppress apoptosis of glioma cells and might serve as a potential oncogene for gliomas.PMID:34080964 | DOI:10.2174/1568009621666210602164225

Cancers, Vol. 13, Pages 3165: The Identification of RNA-Binding Proteins Functionally Associated with Tumor Progression in Gastrointestinal Cancer
a Mikihiro Fujiya Previous investigations have indicated that RNA-binding proteins (RBPs) are key molecules for the development of organs, differentiation, cell growth and apoptosis in cancer cells as well as normal cells. A bioinformatics analysis based on the mRNA expression and a somatic mutational database revealed the association between aberrant expression/mutations of RBPs and cancer progression. However, this method failed to detect functional alterations in RBPs without changes in the expression, thus leading to false negatives. To identify major tumor-associated RBPs, we constructed an siRNA library based o...

DCLK1 Inhibition Sensitizes Colorectal Cancer Cells to Radiation Treatment
Int J Mol Cell Med. 2021 Winter;10(1):23-33. doi: 10.22088/IJMCM.BUMS.10.1.23. Epub 2021 May 22.ABSTRACTColorectal cancer (CRC) is one of the most prevalent diagnosed cancers and a common cause of cancer-related mortality. Despite effective clinical responses, a large proportion of patients undergo resistance to radiation therapy. Therefore, the identification of efficient targeted therapy strategies would be beneficial to overcome cancer radioresistance. Doublecortin-like kinase 1 (DCLK1) is an intestinal and pancreatic stem cell marker that showed overexpression in a variety of cancers. The transfection of DCLK1 siRNA to...

Simultaneous blockade of TIGIT and HIF-1 α induces synergistic anti-tumor effect and decreases the growth and development of cancer cells
CONCLUSIONS: Present data suggest the combination treatment of TIGIT and HIF-1α as a novel treatment strategy against colorectal and breast cancer, but more researches are required to realize the complete role of TIGIT and HIF-1α inside the TME.PMID:34710844 | DOI:10.1016/j.intimp.2021.108288

Late Breaking Abstract - DCLK1 mediates thrombin-induced IL-8/CXCL8 expression in lung epithelial cells
Airway neutrophilia has been known correlated with asthma severity. Thrombin plays a crucial role in asthma. Thrombin can induce IL-8/CXCL8 expression, a potent chemoattractant for neutrophils, in lung epithelial cells. Doublecortin-like kinase 1 (DCLK1) is a protein kinase that has been identified as a tumor stem cell marker in colon and pancreatic cancer. Here we investigated the role of DCLK1 in thrombin-induced IL-8/CXCL8 expression in human lung epithelial cells. Thrombin-induced IL-8/CXCL8 release was attenuated by DCLK1 siRNA or LRKK2-IN-1 (DCLK1 inhibitor) in BEAS-2B and A549 cells. Moreover, thrombin-induced IL-8/...

Auriculasin enhances ROS generation to regulate colorectal cancer cell apoptosis, ferroptosis, oxeiptosis, invasion and colony formation
Biochem Biophys Res Commun. 2021 Dec 1;587:99-106. doi: 10.1016/j.bbrc.2021.11.101. Online ahead of print.ABSTRACTColorectal cancer (CRC) is one of the most common malignant tumors in the digestive system, and Chinese herbal medicine plays an important role in tumor treatment. The in-depth study of auriculasin isolated from Flemingia philippinensis showed that auriculasin promoted reactive oxygen species (ROS) generation in a concentration-dependent manner; when ROS scavenger NAC was added, the effects of auriculasin in promoting ROS generation and inhibiting cell viability were blocked. Auriculasin induced CRC cell apopto...
