Early depletion of M1 macrophages retards the progression of glucocorticoid-associated osteonecrosis of the femoral head
In conclusion, our study reveals the crucial involvement of M1 macrophages in the pathogenesis of GA-ONFH, highlighting their indispensable role in disease progression. Furthermore, early clearance emerges as a promising strategy for impeding the development of GA-ONFH.PMID:37481850 | DOI:10.1016/j.intimp.2023.110639

Gene editing with 'pencil' rather than 'scissors' in human pluripotent stem cells
Stem Cell Res Ther. 2023 Jun 20;14(1):164. doi: 10.1186/s13287-023-03394-5.ABSTRACTOwing to the advances in genome editing technologies, research on human pluripotent stem cells (hPSCs) have recently undergone breakthroughs that enable precise alteration of desired nucleotide bases in hPSCs for the creation of isogenic disease models or for autologous ex vivo cell therapy. As pathogenic variants largely consist of point mutations, precise substitution of mutated bases in hPSCs allows researchers study disease mechanisms with "disease-in-a-dish" and provide functionally repaired cells to patients for cell therapy. To this e...

Notch signaling sculpts the stem cell niche
Front Cell Dev Biol. 2022 Dec 20;10:1027222. doi: 10.3389/fcell.2022.1027222. eCollection 2022.ABSTRACTAdult stem cells depend on their niches for regulatory signaling that controls their maintenance, division, and their progeny differentiation. While communication between various types of stem cells and their niches is becoming clearer, the process of stem cell niche establishment is still not very well understood. Model genetic organisms provide simplified systems to address various complex questions, for example, how is a stem cell niche formed? What signaling cascades induce the stem cell niche formation? Are the mecha...

Cancers, Vol. 14, Pages 4586: Employment among Childhood Cancer Survivors: A Systematic Review and Meta-Analysis
In conclusion, this systematic review and meta-analysis provides evidence that two-thirds of CCS are employed worldwide. Identifying vulnerable groups of CCS may allow for the design of multidisciplinary support strategies and interventions to promote employment in this population.

Generation of Hepatocyte Organoids from Human iPS Cells
Methods Mol Biol. 2022;2544:51-70. doi: 10.1007/978-1-0716-2557-6_3.ABSTRACTHuman-induced pluripotent stem cells (hiPSCs) constitute a great source to generate specialized cells that can be employed in cell replacement therapy for a number of degenerative diseases. In this chapter, I describe a strategy to mass-produce fully functional hepatocyte organoids using hiPSCs interlaced with human adipose microvascular endothelial cells (HAMEC). Our unique technology employs a two-step strategy, consisting of the scalable generation of nearly spherical uniform-sized human embryoid bodies (hEBs), and the subsequent employment of a...

Interferon- γ enhances the immunosuppressive ability of canine bone marrow-derived mesenchymal stem cells by activating the TLR3-dependent IDO/kynurenine pathway
CONCLUSION: Our findings substantiate that IFN-γ stimulation can enhance the immunomodulatory properties of cBMSCs by promoting TLR3-dependent activation of the IDO/kynurenine pathway, increasing the secretion of immunoregulatory molecules and strengthening interactions with T lymphocytes, which may provide a meaningful strategy for the clinical application of cBMSCs in immune-related diseases.PMID:35690960 | DOI:10.1007/s11033-022-07648-y

Photobiomodulation effects on periodontal ligament stem cells: A systematic review of in-vitro studies
CONCLUSIONS: Photobiomodulation can enhance the stemness and differentiation capacities of periodontal ligament stem cells. Therefore, for PBM protocols, there remains no consensus amongst the scientific community. Statistical analyses performed here indicated that the employment of a near-infrared (NIR) wavelength of 940 nm may not yield a significant favourable outcome, although those within the 630 - 830 nm range did so. Concerning the fluence, it should not exceed 8 J/cm2 when therapy is applied by LED devices, and 4 J/cm2 when applied by lasers respectively.PMID:35638280 | DOI:10.2174/1574888X17666220527090321

The Combination of Autologous Mesenchymal Stem cell-derived Exosomes and Neurotrophic Factors as an Intervention for Amyotrophic Lateral Sclerosis
Ann Anat. 2022 Mar 9:151921. doi: 10.1016/j.aanat.2022.151921. Online ahead of print.ABSTRACTAmyotrophic lateral sclerosis is a chronic progressive degeneration of motor neurons and has a high mortality. Riluzole and edaravone are the only approved medications currently being used for amyotrophic lateral sclerosis in clinical settings. However, they can lead to serious complications, such as injuries to the liver and kidney. To date, there is no effective treatment for amyotrophic lateral sclerosis. In this regard, investigations concerning the employment of exosomes, mesenchymal stem cells, and neurotrophic factors to ame...
