Anti-tumor activity of trimethoprim-sulfamethoxazole against melanoma skin cancer through triggering allergic reaction and promoting immunity
This study examined the potential roles of TMP-SMX in melanoma skin cancer using an immunodeficient mouse model. TMP-SMX significantly improved the survival rate and reduced the tumor weight and growth and vascular endothelial growth factor levels in melanoma skin cancer of immunodeficient mice. In the forced swimming test, TMP-SMX significantly reduced immobility time compared to the melanoma skin cancer of immunodeficient mice, indicating improved immunity. TMP-SMX significantly increased infiltration of mast cells and release of allergy-related mediators (IgE, histamine, interleukin (IL)-4, IL-5, IL-13, and IL-33) and i...

Drug allergy biomarkers
Although the true burden of drug allergy is unknown, several studies indicate that adverse drug reactions occur in up to 6% to 10% of hospitalized patients —20% of which are because of drug allergy—and up to 10% of the general population claims to have penicillin or other drug allergies.1 A plethora of new antibiotics, vaccines, chemotherapy drugs, and, more recently, biologicals and targeted molecular drugs are available and have improved disease treatment options, but drug allergy remains a vexing and unaddressed problem that deprives patients of the best medication for their disease, and when re-exposed, places them...

Drug allergy desensitization is not a unique recipe
Purpose of review Drug desensitization is the only therapeutic option for patients with drug allergies who need to receive the drugs they are allergic to, and it is especially critical in patients with an urgent need for chemotherapy, biologics, or antibiotics, where equally effective alternatives might not be available. However, drug desensitization is not a cookbook where anyone with no experience or specific training can find a general recipe. This review article will approach the singularities that make personalized and highly specialized care essential in this field. Recent findings Drug desensitization nee...

Guideline on diagnostic procedures for suspected hypersensitivity to beta-lactam antibiotics
AbstractThis guideline on diagnostic procedures for suspected beta-lactam antibiotic (BLA) hypersensitivity was written by the German and Austrian professional associations for allergology, and the Paul-Ehrlich Society for Chemotherapy in a  consensus procedure according to the criteria of the German Association of Scientific Medical Societies. BLA such as penicillins and cephalosporins represent the drug group that most frequently triggers drug allergies. However, the frequency of reports of suspected allergy in patient histories cle arly exceeds the number of confirmed cases. The large number of suspected BLA allergies ...

Indeed, they can recognize molecules induced at the cell surface by stress signals...

Evaluation of Morbidity and Mortality from Drug Allergies in Patient with Acute Myeloid Leukemia
Patients with a drug allergy are known to have increased morbidity and mortality. Patients with Acute Myeloid Leukemia (AML) are known to have recurrent severe infections due to immunosuppression from chemotherapy. The prevalence of drug allergy in patients with AML and its effect on morbidity and mortality has not been previously studied.

Options in Hypersensitivity Reactions to Chemotherapeutics
This article has tried to highlight and summarize the most recent scientific progress concerning risk factors, pathogenesis, diagnosis, and treatment of these reactions.Recent findingsIdentification of patients at high risk of developing hypersensitivity reactions allows risk stratification to guide clinical decision-making. Therefore, the most recent researches evaluated the possibility to perform risk stratification in case of hypersensitivity reactions to platinum compounds and taxanes. In addition, new data are now available regarding the role of in vitro test for the diagnosis of reactions to platins and the role of d...

Desensitization for hypersensitivity reactions to medications.
Authors: del Carmen Sancho M, Breslow R, Sloane D, Castells M Abstract Rapid drug desensitization (RDD) is a technique that induces temporary tolerance to a drug, allowing a medication-allergic patient to receive the optimal agent for his or her disease. Through RDD, patients with IgE and non-IgE hypersensitivity reactions (HSRs) including anaphylaxis can safely be administered important medications while minimizing or completely inhibiting adverse reactions. Adverse reactions to drugs are increasingly recognized as important contributors to disease as well as impediments to the best treatment of dermatological, in...

Combined DOCK8 and CLEC7A mutations causing immunodeficiency in 3 brothers with diarrhea, eczema, and infections
We report 4 brothers from a consanguineous Italian family with an immunodeficiency syndrome characterized by severe eczema, milk and egg allergies, recurrent infections, intractable diarrhea, failure to thrive, and, in 2 of the 3 surviving brothers, lymphoma. One of the brothers (patient II-2) died at age 12 months of cardiovascular failure caused by intractable diarrhea, and the other 3 were evaluated as part of this study ( and , A). The proband (patient II-1) presented shortly after birth with severe eczema and intractable diarrhea without gastrointestinal infection. In the first year of life, he had multiple allergies ...
