Comparative outcomes associated with rivaroxaban versus warfarin use in elderly patients with atrial fibrillation or acute venous thromboembolism managed in the United States: a systematic review of observational studies
CONCLUSIONS: This systematic review supports findings from subgroup analyses of randomized controlled trials that, compared with warfarin, rivaroxaban is associated with generally neutral or positive effects on thrombosis and a mixed picture on bleeding outcomes in older adults with either NVAF or VTE treated in the United States.PMID:37584187 | DOI:10.1080/03007995.2023.2247988

Molecular cloning of Reteplase and its expression in E. coli using tac promoter.
CONCLUSION: In the present study, reteplase cDNA was successfully cloned and expressed using tac promoter. This vector will be used for the optimization of the expression of reteplase in E. coli. PMID: 25298959 [PubMed]

Betaine (N,N,N-trimethylglycine) averts photochemically-induced thrombosis in pial microvessels in vivo and platelet aggregation in vitro
In conclusion, our data show that betaine protected against coagulation events in vivo and in vitro and decreased LPO in plasma.

Ixodes scapularis Tissue Factor Pathway Inhibitor
Ixodes scapularis is a blood-sucking tick and the principal vector of Lyme disease, a spirochetal illness caused by Borrelia burgdorferi and now the most common vector-borne infection in the United States; more than 50,000 cases have been reported during the last ten years. The salivary gland of I. scapularis has a number of pharmacologically active molecules that help the tick to successfully feed on blood, such as inhibitors of complement system, in addition to coagulation and platelet aggregation inhibitors. This invention describes Ixolaris, a protein that inhibits the initiation of blood coagulation by inhibition of c...

Comparison of First- and Second-Generation Drug-Eluting Stents in an All-Comer Population of Patients with Diabetes Mellitus (from Katowice-Zabrze Registry).
CONCLUSIONS In a real-life setting of diabetic patients undergoing PCI, second-generation DES had lower risk of ST and similar MACCE rate compared to first-generation DES. PMID: 26506478 [PubMed - as supplied by publisher]

Chronic, not acute, skin-specific inflammation promotes thrombosis in psoriasis murine models
Background: Psoriasis patients exhibit an increased risk of atherothrombotic events, including myocardial infarction and stroke. Clinical evidence suggests that psoriasis patients with early onset and more severe disease have the highest risk for these co-morbidities, perhaps due to the extent of body surface involvement, subsequent levels of systemic inflammation, or chronicity of disease. We sought to determine whether acute or chronic skin-specific inflammation was sufficient to promote thrombosis. Methods: We used two experimental mouse models of skin-specific inflammation generated in either an acute (topical Aldara a...

Mp13: important role of prothrombin time (pt) and partial thromboplastin time (ptt) in predicting tpa-related hemorrhagic transformation
Conclusions Higher PT/PTT level within 72 hr of IV tPA is early marker of tPA-induced HT. Whether these routine labs have value in symptomatic hemorrhage will require further study in a larger cohort. But this proof-of-concept study suggests that tPA efficacy can potentially be followed in real time. The development of a reliable blood test would be of clinical utility to gauge thrombolytic efficacy in real time to guide and triage adjunct treatments. Abstract MP13 Figure 1

Id: 41: thrombotic ramifications of ophthalmologic diagnosis of familial and acquired thrombophilia as pathoetiology for otherwise unexplained uveitis.
Conclusion When the usual pathoetiologies of uveitis are not found, ophthalmologists should screen for thrombophilia, often being the first to illuminate treatable etiologies for other thrombotic events. In our 3 patients, when the pathoetiology of uveitis could not be discerned, screening for familial and acquired thrombophilia had important therapeutic ramifications in the probands and their first degree relatives. .

State transition model: vorapaxar added to standard antiplatelet therapy to prevent thrombosis post myocardial infarction or peripheral artery disease.
CONCLUSION: This study contributes to the growing literature on secondary prevention add-on therapy, as results from these modeling analyses suggest adding vorapaxar to SC for patients at high atherothrombotic risk can provide long-term health benefits. Factor VII activating protease (FSAP) influences vascular remodeling in the mouse hind limb ischemia model.
CONCLUSIONS: Arteriogenesis is enhanced, and this is associated with a higher infiltration of monocytes, in the absence of endogenous FSAP but angiogenesis is unchanged. Exogenous FSAP had the opposite effect on arteriogenesis indicating a possible therapeutic potential of modulating endogenous FSAP. PMID: 28670395 [PubMed]

Prothrombotic Fibrin Clot Phenotype in Patients with Deep Vein Thrombosis and Pulmonary Embolism: A New Risk Factor for Recurrence.
Abstract Prothrombotic fibrin clot phenotype, involving faster formation of dense meshwork composed of thinner and highly branched fibers that are relatively resistant to plasmin-induced lysis, has been reported in patients with not only myocardial infarction or stroke, but also venous thromboembolism (VTE), encompassing deep vein thrombosis (DVT), and/or pulmonary embolism (PE). Prothrombotic fibrin clot phenotype, in particular prolonged clot lysis time, is considered a novel risk factor for VTE as well as venous thrombosis at unusual location, for example, cerebral sinus venous thrombosis, retinal vein obstruct...

Influence of age on the pharmacokinetics, pharmacodynamics, efficacy, and safety of rivaroxaban.
CONCLUSION: A favorable clinical profile with rivaroxaban was observed across age subgroups, supporting the premise that dosing in older adults does not necessitate adjustment. However, it is prudent that a cautious and individualized approach is taken for treatment with any anticoagulant in older adults. PMID: 29932775 [PubMed - as supplied by publisher]

Oral anticoagulation therapy and subsequent risk of venous thromboembolism in atrial fibrillation patients.
CONCLUSIONS: In this large practice-based population of AF patients prescribed OACs for primary prevention of stroke and systemic embolization, subsequent risk of VTE was lowest among those prescribed apixaban and dabigatran, while risk was similar with prescriptions for warfarin and rivaroxaban. Among AF patients prescribed OACs, lowering risk of VTE may be an additional benefit of apixaban and dabigatran, beyond the reduced bleeding risk observed in randomized clinical trials.
