Investigating bevacizumab and its fragments sustained release from intravitreal administrated PLGA Microspheres: A modeling approach
This study discusses certain issues related to the sustained release of bevacizumab from intravitreal poly(lactic-co-glycolic acid) (PLGA) microspheres. A computational model elucidating the ocular kinetics of bevacizumab is demonstrated, wherein the release of the drug from PLGA microspheres is modeled using the Koizumi approach, complemented by an empirical model that links the kinetics of bevacizumab release to a size-dependent hydrolytic degradation of the drug-loaded polymeric microparticles. The results of the simulation were then rigorously validated against experimental data. The as-developed model proved remarkabl...

Oct biomarkers for early prognosis in diabetic macular edema treatment with ranibizumab
CONCLUSION: This study showed the value and importance of OCT parameters, such as absence of INLc, as a prognostic therapeutic response. A normalization of the macular anatomy with ranibizumab is more likely to happen in early complete responders. The association between INLc and higher glycated haemoglobin levels showed the importance of systemic metabolic control in systemic diabetic manifestations. Vitreous Floaters With New Intravitreal Drugs Administered by Retina Specialists
In this issue of JAMA Ophthalmology, Dessouki and colleagues describe symptomatic silicone oil microdroplets in 16 of 55 patients (29%) receiving pegcetacoplan following a single injection using a Luer lock 1-mL syringe sold by McKesson (16-PS1C). It is not unexpected that silicone oil microdroplets would be noted in some eyes following intravitreal injection because they have been reported in multiple studies for eyes receiving bevacizumab, ranibizumab, and aflibercept. The remarkable feature in this study is the high percentage of patients with symptomatic floaters after a single injection of pegcetacoplan because in pri...

Comparative Efficacy, Durability and Safety of Faricimab in the Treatment of Diabetic Macular Edema: A Systematic Literature Review and Network Meta-Analysis
ConclusionFaricimab provides a new treatment option in DME with dual-pathway inhibition of VEGF and angiopoeitin-2 (Ang-2). To the authors ’ knowledge, this is the first indirect comparison of faricimab T&E in DME. The analyses indicate that faricimab T&E is associated with superior retinal drying along with numerically fewer injections compared to all other treatments given in flexible dosing regimens. It also showed superior visual acuity outcomes compared to ranibizumab and bevacizumab. (Source: Advances in Therapy)

Effectiveness Differences of Ranibizumab and Aflibercept Action in the Treatment of Diabetic Retinopathy
CONCLUSION: Both vitreous cavity injections of LU and AF can effectively reduce the expression of vascular-related factors in the atrial fluid of DR patients, but AF has a more significant inhibitory effect on the level of inflammatory factors in patients in the short term after treatment.PMID:37820680 (Source: Alternative Therapies in Health and Medicine)
