A potent bioreducible ionizable lipid nanoparticle enables siRNA delivery for retinal neovascularization inhibition
Eur J Pharm Biopharm. 2024 Apr 16:114296. doi: 10.1016/j.ejpb.2024.114296. Online ahead of print.ABSTRACTSmall interfering RNA (siRNA) is emerging as a promising treatment for retinal neovascularization due to its specific inhibition of the expression of target genes. However, the clinical translation of siRNA drugs is hindered by the efficiency and safety of delivery vectors. Here, we describe the properties of a new bioreducible ionizable lipid nanoparticle (LNP) 2N12H, which is based on a rationally designed novel ionizable lipid called 2N12B. 2N12H exhibited degradation in response to the mimic cytoplasmic glutathione ...

Exploring the Therapeutic Potential of Anti-VEGF Drugs for the Management of Diabetic Retinopathy: An Overview
Curr Diabetes Rev. 2024 Apr 16. doi: 10.2174/0115733998294968240405091023. Online ahead of print.ABSTRACTThe discovery of antivascular endothelial growth factor medications has resulted in a substantial change in diabetic retinopathy treatment. The most common cause of diabetic retinopathy blindness is Diabetic Macular Edema. The pathophysiology of Diabetic Macular Edema is thought to include the well-known pro-angiogenic and pro-permeability factor vascular endothelial growth factor. Over the past decade, drugs that impede the functions of vascular endothelial growth factors have established themselves as a standard-of-ca...

Cognitive bias evaluation on the choice of treatment in common retinal disorders among retina specialists in 2023
CONCLUSION: The findings regarding the first and second line choices for the mentioned ocular disorders were found to agree with the findings of published literature currently used in practice, with a tendency to prefer ranibizumab as first line therapy for neovascular disorders and aflibercept as first line therapy for macular edema. In addition, there were no differences between choices for first and second line therapy for patients vs. ophthalmologists.PMID:38603895 | DOI:10.1016/j.jfo.2024.104177 (Source: Journal Francais d Ophtalmologie)

Comparative quantitative analysis of optical coherence tomography angiography in varied morphologies of macular neovascularization following intravitreal conbercept and ranibizumab treatments for neovascular age ‑related macular degeneration
Exp Ther Med. 2024 Mar 20;27(5):214. doi: 10.3892/etm.2024.12501. eCollection 2024 May.ABSTRACTThe present study aimed to examine the optical coherence tomography angiography (OCTA) parameters associated with macular neovascularization (MNV) in patients diagnosed with neovascular age-related macular degeneration (nAMD) and treated with either intravitreal conbercept (IVC) or ranibizumab (IVR). It enrolled 39 nAMD patients presenting with MNV, including 23 in the IVC group and 16 in the IVR group. All participants were treatment-naïve with intravitreal therapy and they underwent treatment following a '3+PRN' regimen. The M...

Molecular Basis of Angiogenesis and its Application
Keio J Med. 2024;73(1):12. doi: 10.2302/kjm.ABSTRACT_73_1-1.ABSTRACTAngiogenesis, the development of new blood vessels, is a fundamental physiological process. In addition, angiogenesis plays a key role in the pathogenesis of several disorders, including cancer and eye disorders such as diabetic retinopathy and age-related macular degeneration (AMD). However, identifying the regulators of angiogenesis proved challenging. Numerous factors that stimulated angiogenesis in various bioassays were identified, but their pathophysiological role remained unclear. In 1989, we reported the isolation and cloning of vascular endothelia...
