Formulation and investigation of hydrogels containing an increased level of diclofenac sodium using risk assessment tools
Eur J Pharm Sci. 2023 Dec 9:106666. doi: 10.1016/j.ejps.2023.106666. Online ahead of print.ABSTRACTTransdermal delivery of active ingredients is a challenge for pharmaceutical technology due to their inadequate penetration properties and the barrier function of the skin. The necessity of painless, effective, topical therapy for the aging population is growing, and a variety of diclofenac sodium-containing semi-solid preparations are available to alleviate the symptoms of these ailments. Our purpose was to formulate a novel composition with higher drug content to enhance drug release and permeation, thereby providing more e...

Population pharmacokinetics of methotrexate and 7-hydroxymethotrexate and delayed excretion in infants and young children with brain tumors
CONCLUSIONS: MTX and 7OHMTX plasma disposition were characterized for the first time in young children with brain tumors. No exposure-toxicity relationship was identified in this study, presumably due to aggressive clinical management which led to a low MTX-related toxicity rate.PMID:38070781 | DOI:10.1016/j.ejps.2023.106669 (Source: European Journal of Pharmaceutical Sciences)

Infliximab aggregated produced in severe and mild elevated temperature stress conditions induce an extended specific CD4 T-cell response
Eur J Pharm Sci. 2023 Dec 7:106670. doi: 10.1016/j.ejps.2023.106670. Online ahead of print.ABSTRACTAggregation has been widely described as a factor contributing to therapeutic antibody immunogenicity. Although production of high-affinity anti-drug antibodies depends on the activation of CD4 T lymphocytes, little is known about the T-cell response induced by antibody aggregates, especially for aggregates produced in mild conditions resulting from minor handling errors of vials. Large insoluble infliximab (IFX) aggregates produced in severe elevated temperature stress conditions have been previously shown to induce human mo...

In vitro trypanocidal potency and in vivo treatment efficacy of oligomeric ethylene glycol-tethered nitrofurantoin derivatives
Eur J Pharm Sci. 2023 Dec 7;192:106668. doi: 10.1016/j.ejps.2023.106668. Online ahead of print.ABSTRACTAfrican trypanosomiasis is a significant vector-borne disease of humans and animals in the tsetse fly belt of Africa, particularly affecting production animals such as cattle, and thus, hindering food security. Trypanosoma congolense (T. congolense), the causative agent of nagana, is livestock's most virulent trypanosome species. There is currently no vaccine against trypanosomiasis; its treatment relies solely on chemotherapy. However, pathogenic resistance has been established against trypanocidal agents in clinical use...

Preparation of paeoniflorin-glycyrrhizic acid complex transethosome gel and its preventive and therapeutic effects on melasma
Eur J Pharm Sci. 2023 Dec 5:106664. doi: 10.1016/j.ejps.2023.106664. Online ahead of print.ABSTRACTPaeoniflorin (PF) and glycyrrhizic acid (GL) have skin beautifying effects of anti-inflammation, anti-oxidation, inhibition of melanin formation, and reduction of skin pigmentation. To improve the transdermal permeability of PF and GL in transdermal drug delivery system (TDDS) and enhance their anti-melasma efficacy, PF-GL transethosome (PF-GL-TE) was prepared by ethanol injection method, and finally gelled with carbomer-940 to form PF-GL-TE gel. Consequently, the obtained PF-GL-TE is small and uniform, with an average partic...

Folic acid-functionalized chitosan nanoparticles with bioenzyme activity for the treatment of spinal cord injury
Eur J Pharm Sci. 2023 Dec 6;192:106667. doi: 10.1016/j.ejps.2023.106667. Online ahead of print.ABSTRACTSpinal cord injury (SCI) is a central system disease with a high rate of disability. Pathological changes such as ischemia and hypoxia of local tissues, oxidative stress and apoptosis could lead to limb pain, paralysis and even life-threatening. It was reported that catalase (CAT) was the main antioxidant in organisms, which could remove reactive oxygen species (ROS) and release oxygen (O2). However, the efficacy of the drug is largely limited due to its poor stability, low bioavailability and inability to cross the blood...

Formulation Development of Pharmaceutical Nanoemulgel for Transdermal Delivery of Feboxostat: Physical Characterization and In vivo Evaluation
This study aimed to fabricate and characterize feboxostat (FXT) loaded nanoemulgel (NEG) for transdermal delivery. NEG was prepared by high sheared homogenization technique and characterized for thermodynamic stability, pH analysis, drug content, zeta analysis, viscosity, spreadability, FTIR, in-vitro drug release and ex-vivo permeation. In vivo anti-inflammatory activity was evaluated in albino rats by inducing edema in hind paws using carrageenan. The formulations showed optimum thermodynamic stability, having no phase separation and color change. The pH was in the range of human skin range i.e. 5.5-6.5. The drug content...

A First-in-Human Study of Brozopentyl Sodium, following Single and Multiple Ascending Intravenous Infusion in Chinese Healthy Volunteers
CONCLUSION: BZP displayed safety, good tolerability and predictable PK characteristics following both single and multiple ascending intravenous administrations. These findings provide a basis for further clinical development of BZP for ischemic stroke patients.PMID:38056780 | DOI:10.1016/j.ejps.2023.106663 (Source: European Journal of Pharmaceutical Sciences)
