Recent advances and clinical translation of liposomal delivery systems in cancer therapy
We describe the recent understanding of spatiotemporal fate of liposomes in the organism after different routes of drug administration. Meanwhile, various types of liposome-based drug delivery systems that exert their respective advantages in cancer therapy while reducing side effects were discussed. Moreover, the combination of liposomal agents with other therapies (e.g. photodynamic therapy; photothermal therapy, etc.) which allows for higher tumor-targeting efficiency and therapeutic efficacy was systematically evaluated. Finally, the opportunities and challenges faced by the field of liposome nanoformulations for enter...

Cancer sensitizing effect of deazaflavin analogs is associated with increased intracellular drug accumulation
Eur J Pharm Sci. 2023 Dec 28:106686. doi: 10.1016/j.ejps.2023.106686. Online ahead of print.ABSTRACTAs part of our efforts geared towards developing mechanism-based cancer sensitizing agents, we have previously synthesized and characterized novel deazaflavin analogs as potent tyrosyl DNA phosphodiesterase 2 (TDP2) inhibitors for combination treatments with topoisomerase II (TOP2) poisons. Interestingly, the sensitizing effect of a few analogs toward TOP2 poison etoposide (ETP) was associated with a significant increase in intracellular drug accumulation, which could be an alternative mechanism to boost the clinical efficac...

Assessing the performance of new chromatographic technologies for the separation of peptide epimeric impurities: the case of Icatibant
Eur J Pharm Sci. 2023 Dec 22:106682. doi: 10.1016/j.ejps.2023.106682. Online ahead of print.ABSTRACTThe biopharmaceutical industry faces the challenge of efficiently characterising impurity profiles of therapeutical peptides, also due to their complex polar and ionisable attributes. This research explores the potential of advanced chromatographic techniques to address this challenge. The study compares dynamic electrostatic repulsion reversed phase (d-ERRP) to its counterparts (static ERRP and ion pair reversed phase IP-RP) in analysing Icatibant and its elusive epimeric impurity, [L-Arg]1-Icatibant and highlights its exce...
