Pharmacological insights on novel oral selective estrogen receptor degraders in breast cancer
Eur J Pharmacol. 2024 Feb 23:176424. doi: 10.1016/j.ejphar.2024.176424. Online ahead of print.ABSTRACTThe therapeutic landscape of estrogen receptor (ER)-positive breast cancer includes endocrine treatments with aromatase inhibitors (AIs), selective estrogen receptor modulators (SERMs), and selective estrogen receptor degraders (SERDs). Fulvestrant is the first approved SERD with proven efficacy and good tolerability in clinical practice. However, secondary drug resistance, low receptor affinity, and parental administration stimulated the search for new oral SERDs opening a new therapeutic era in ER + breast cancer. Elaces...

Concurrent factors associated with adherence to adjuvant endocrine therapy among women with non-metastatic breast cancer
ConclusionsMedication-taking self-efficacy and employment status were associated with AET adherence above other related factors.Implications for Cancer SurvivorsEnhancing patients ’ confidence in their ability to take AET for breast cancer may represent an important intervention target to boost adherence. Machine learning predicts the risk of osteoporosis in patients with breast cancer and healthy women
CONCLUSIONS: Patients with breast cancer may have changed body composition and BMD. Compared with the healthy population, the main indicators of osteoporosis in patients with breast cancer were reduced nonadipose tissue, increased risk of edema, altered fat distribution, and reduced BMD. In addition to age, duration of treatment, and duration of menopause, body composition-related indicators such as BMI and basal metabolism may be considerably associated with BMD of patients with breast cancer, suggesting that BMD status can be monitored in clinical practice by focusing on changes in the aforementioned indexes, which may p...

GSE251644 Cancer-associated fibroblasts rewire the estrogen receptor response in luminal breast cancer, enabling estrogen independence
In conclusion, our work reveals that CAFs directly control the luminal breast cancer phenotype by selectively modulating ER-α expression and transcriptional function, and further proposes novel targ ets to disrupt the crosstalk between CAFs and tumor cells to reinstate treatment response to endocrine therapy in patients. Recommendations for cyclin ‑dependent kinase 4/6 inhibitor treatments in the context of co‑morbidity and drug interactions (Review)
Oncol Lett. 2024 Feb 8;27(4):145. doi: 10.3892/ol.2024.14278. eCollection 2024 Apr.ABSTRACTBreast cancer is most frequently diagnosed among women aged 65-74 years and the prevalence of comorbidities in elderly patients with breast cancer is 32.2%. In addition, polypharmacy is quite common in these patients. Understanding the interaction between breast cancer treatment modalities and comorbidities is important, particularly in elderly patients, as comorbidities affect the choice of appropriate treatment and are independent risk factors for survival. A total of three oral cyclin-dependent kinase 4 and 6 inhibitors (CDK4/6i),...
