Viruses, Vol. 13, Pages 1918: Application Route and Immune Status of the Host Determine Safety and Oncolytic Activity of Oncolytic Coxsackievirus B3 Variant PD-H Viruses doi: 10.3390/v13101918 Authors: Ahmet Hazini Babette Dieringer Karin Klingel Markian Pryshliak Anja Geisler Dennis Kobelt Ole Daberkow Jens Kurreck Sophie van Linthout Henry Fechner The coxsackievirus B3 strain PD-0 has been proposed as a new oncolytic virus for the treatment of colorectal carcinoma. Here, we generated a cDNA clone of PD-0 and analyzed the virus PD-H, newly generated from this cDNA, in xenografted and syngenic models of colorectal cancer. Replication and cytotoxic assays revealed that PD-H replicated and lysed colorectal carcinoma cell lines in vitro as well as PD-0. Intratumoral injection of PD-H into subcutaneous DLD-1 tumors in nude mice resulted in strong inhibition of tumor growth and significantly prolonged the survival of the animals, but virus-induced systemic infection was observed in one of the six animals. In a syngenic mouse model of subcutaneously growing Colon-26 tumors, intratumoral administration of PD-H led to a significant reduction of tumor growth, the prolongation of animal survival, the prevention of tumor-induced cachexia, and the elevation of CD3+ and dendritic cells in the tumor microenvironment. No virus-induced side effects were observed. After intraperitoneal application, PD-H induced weak pancreatitis and myocarditis in immunocompetent mic...

ConclusionsImprovements in the detection of HCV combined with a simplified one-stop testing and diagnostic approach represents an opportunity to reduce the burden of HCV in Iraq and may play a significant role in meeting World Health Organisation HCV elimination targets.

Aging is built on feedback loops, interactions between damage and dysfunction in which both sides accelerate the other. This is a feature of all complex systems, not just biological ones. The existence of feedback loops in which damage accumulation causes dysfunction that accelerates damage accumulation is the fundamental reason as to why aging is an accelerating process. It starts slowly, and moves ever more rapidly with time. Aging in one's 30s is a very different beast to aging in one's 70s, and the downhill slide is much faster in later life. The way to break this cycle is to repair the damage. This is true whet...
Meeting of the Working Group on the National Mental Health Strategic Plan, April 2019By Saima Wazed and Nazish ArmanDHAKA, Oct 27 2021 (IPS) Mental health is a state of well-being when both your body and your mind are in balance, and you are able to deal with the difficulties and challenges that come your way and easily find joy, peace, and happiness once the challenge is overcome. For many people though, the challenges often remain for too long – the pain of losing someone you dearly loved, being diagnosed with a chronic disease like cancer or a heart condition, losing your family/home/job or feeling like you failed...
