Experimental dapsone administration induces infertility in male Wistar rats: mechanisms and clinical implications

This study investigated the effect of the administration of dapsone on the reproductive activities of male rats using in vivo and in vitro techniques. In the in vivo study, dapsone was administered orally to male Wistar rats for 5 days or 6 weeks after which their body weight, relative reproductive organ weights, sperm parameters and reproductive hormones were determined while testicular and epididymal histology were also assessed. Data were compared using analysis of variance and Students-Newman-Keuls multiple comparison test. For the in vitro study, Sertoli cells were cultured and treated with varying doses of dapsone at different durations, thereafter Sertoli cell viability and nuclei integrity were determined. Also, the genetic expressions of Glial cell line-derived neurotrophic factor (GDNF) and transferrin were assessed. The results obtained from the in vivo study showed a duration-dependent significant decrease in body and reproductive organ weights, sperm parameters and serum testosterone concentration. Testicular and epididymal histology also showed duration-dependent degenerative changes. However, all these changes were restored towards control values in the recovery experiment. The viability and deoxyribonucleic acid (DNA) integrity of the treated Sertoli cells showed dose and duration-dependent adverse effects while GDNF and transferrin showed normal genetic expressions. These results suggest that dapsone could induce male reproductive stress by affecting testicul...

Hunt, Sierra Goldsmith, Yuting Fan, Sherman J. Silber, Amander T. ClarkAbstractWe generated three human induced pluripotent stem cell (hiPSC) sublines from human dermal fibroblasts (HDF) (MZT04) generated from a skin biopsy donated from a fertile woman. The skin biopsy was broadly consented for generating hiPSC lines for biomedical research, including unique consent specifically for studying human fertility and infertility. hiPSCs were reprogrammed using Send...

When patients with lupus are hospitalized, they should be screened and likely treated for venous thromboembolism, researchers of a nationwide study say. In May, ACR Open Rheumatology published results of the large retrospective study spanning several years. Researchers analyzed trends in mortality, morbidity and hospitalization from venous thromboembolism (VTE), specifically among patients diagnosed with systemic... [Read More]

This study investigated the anti-allergic effect of Poncirus trifoliata (L.) Raf. (PT) on human keratinocytic HaCaT cells in vitro and on 2,4-dinitrochlorobenzene (DNCB)-induced atopic dermatitis-like lesions in vivo. The release of TARC, MCP-1, IL-6 and IL-8 is increased by IFN-γ and TNF-α in HaCaT cells, and PT extract suppressed the increased production of TARC, MCP-1, IL-6, and IL-8. PT extract recovered the expression of filaggrin decreased by IFN-γ and TNF-α. in vivo experiment, PT administration decreased the skin severity score, thickening of the epidermis, movement of inflammatory cells int...

Sjögren's syndrome in Systemic Lupus Erythematosus - a subset characterized by a systemic inflammatory state. J Rheumatol. 2019 Sep 15;: Authors: Ruacho G, Kvarnström M, Zickert A, Oke V, Rönnelid J, Eketjäll S, Elvin K, Gunnarsson I, Svenungsson E Abstract OBJECTIVE: Secondary Sjögren's syndrome (SLE-sSS) is an often-neglected subset of patients with Systemic lupus erythematosus (SLE). Furthermore, primary Sjögren's syndrome overlaps and can be difficult to delineate from SLE. To shed light on the SLE-sSS subset, we investigated a large and well-characterized SLE cohort c...
