Modulation of the autophagy ‐lysosomal pathway and endoplasmic reticulum stress by ketone bodies in experimental models of stroke
We report for the first time that the protective effect of β-hydroxybutyrate (BHB) against stroke and glucose deprivation is enantiomer selective. We discovered that D-BHB treatment prevented the cleavage of the lysosomal membrane protein LAMP2, maintained lysosome integrity and stimulated the autophagic flux. Also, D-BHB notably reduced the activation of the PERK and IRE1α branches of the unfolded protein response (UPR), attenuating protein synthesis inhibition and preventing endoplasmic reticulum stress. Together our results support the therapeutic use of D-BHB treatment post-ischemia, by preventing the loss of proteos...

Television Viewing Time and All-cause and Cardiovascular Disease Mortality Among Japanese Adults with and without a History of Stroke or Myocardial Infarction
CONCLUSIONS: Prolonged TV viewing time was associated with higher risks of all-cause and CVD mortality in stroke or MI survivors and in persons without a history of them. It may be recommended to reduce sedentary time for stroke or MI survivors, independent of the level of physical activity.PMID:37225520 | DOI:10.5551/jat.63959

Heterozygosity for ADP-ribosylation factor 6 suppresses the burden and severity of atherosclerosis
PLoS One. 2023 May 10;18(5):e0285253. doi: 10.1371/journal.pone.0285253. eCollection 2023.ABSTRACTAtherosclerosis is the root cause of major cardiovascular diseases (CVD) such as myocardial infarction and stroke. ADP-ribosylation factor 6 (Arf6) is a ubiquitously expressed GTPase known to be involved in inflammation, vascular permeability and is sensitive to changes in shear stress. Here, using atheroprone, ApoE-/- mice, with a single allele deletion of Arf6 (HET) or wildtype Arf6 (WT), we demonstrate that reduction in Arf6 attenuates atherosclerotic plaque burden and severity. We found that plaque burden in the descending...

Ischaemic stroke with multi-focal venous and arterial thrombosis due to hyperhomocysteinemia: anabolic androgenic steroid use and MTHFR c.667 C   >  T variant – a case report
Severely elevated serum homocysteine is a rare cause of ischaemic stroke and extra-cranial arterial and venous thrombosis. Several factors can lead to mild elevation of homocysteine including dietary folate an...

Inflammation May Be the Culprit Behind Our Deadliest Diseases
In the early days of my medical residency, I met a man whom we’ll call Jason. He arrived to our emergency room on a holiday, nonchalant yet amiable, and complained of mild chest pain. Jason was tall and trim, with a strong South Boston accent and fingertips still faintly stained from his last home-improvement project. He was only 45 years old, but he looked much younger. He didn’t smoke, barely drank alcohol, and his cholesterol levels had always been normal. No one in his family had a history of heart disease. He asked us if we could work quickly—he wanted to be home for dinner with his daughters. [time-...

Stroke-induced hexokinase 2 in circulating monocytes exacerbates vascular inflammation and atheroprogression
CONCLUSIONS: Stroke-induced monocyte Hk2 upregulation is a key mechanism underlying post-stroke vascular inflammation and atheroprogression.PMID:36893911 | DOI:10.1016/j.jtha.2023.02.021

The role of gut-dependent molecule trimethylamine N-oxide as a novel target for the treatment of chronic kidney disease
Int Urol Nephrol. 2023 Feb 16. doi: 10.1007/s11255-023-03500-9. Online ahead of print.ABSTRACTTrimethylamine N-oxide (TMAO) is an intestinal uremic toxin molecule mainly excreted by the kidney. Therefore, the plasma TMAO concentration is significantly increased in chronic kidney disease (CKD) patients, and plasma TMAO can be cleared by dialysis. Furthermore, TMAO damage the kidney mainly through three mechanisms: oxidative stress, inflammation and endoplasmic reticulum stress. Clinical experiments have indicated that higher TMAO levels are strongly related to the elevated incidence and mortality of cardiovascular (CV) even...

Endothelial Damage Arising From High Salt Hypertension Is Elucidated by Vascular Bed Systematic Profiling
CONCLUSIONS: These findings provide novel molecular insight into HS-induced structural changes in eGC and EC composition that may increase cardiovascular risk and potentially guide the development of new diagnostics and therapeutic interventions.PMID:36700429 | DOI:10.1161/ATVBAHA.122.318439
