Fluorescent peptide biosensor for monitoring CDK4/cyclin D kinase activity in melanoma cell extracts, mouse xenografts and skin biopsies
Publication date: 15 November 2016 Source:Biosensors and Bioelectronics, Volume 85 Author(s): Camille Prével, Morgan Pellerano, Juan A. González-Vera, Pauline Henri, Laurent Meunier, Julien Vollaire, Véronique Josserand, May C. Morris Melanoma constitutes the most aggressive form of skin cancer, which further metastasizes into a deadly form of cancer. The p16INK4a-Cyclin D-CDK4/6-pRb pathway is dysregulated in 90% of melanomas. CDK4/Cyclin D kinase hyperactivation, associated with mutation of CDK4, amplification of Cyclin D or loss of p16INK4a leads to increased risk of developing melanoma. This kinase therefo...

Targeting mitosis-regulating genes in cisplatin-sensitive and -resistant melanoma cells: A live-cell RNAi screen displays differential nucleus-derived phenotypes.
Abstract Chemoresistance in malignant melanoma remains an unresolved clinical issue. In the search for novel molecular targets a live-cell high-content RNAi screen based on gene expression data was performed in cisplatin-sensitive and cisplatin-resistant MeWo melanoma cells, Mel-28 cells and a melanocyte cell line. Cells were exposed to 91 siRNAs and distinct nucleus-derived phenotypes such as cell division, cell death and migration phenotypes were detected by time-lapse microscopy over 60 hours. Using this approach, cisplatin-sensitive and cisplatin-resistant melanoma cells were compared by automated image analys...

Effect of small interfering RNAs on matrix metalloproteinase 1 expression
Publication date: December 2014 Source:Biotechnology Reports, Volume 4 Author(s): Gen-Hung Chen , Chun-Hua Huang , Huang-Yao Luo , Shann-Tzong Jiang Three small double strand siRNAs (506-MMP1, 859-MMP1 and 891-MMP1), each contains 25–26 nucleotides, with high specific to human MMP1 were designed according to mRNA sequence of human MMP1 (NCBI, NM_002421). To monitor the MMP1 gene expression, the total RNAs of human skin fibroblast (Detroit 551, BCRC 60118) were extracted. One human matrix metalloproteinase 1 (MMP1) partial sequence cDNA, included all the three siRNA target sequences, amplified specifically via RT-PCR an...

Efficient in vitro gene therapy with PEG siRNA lipid nanocapsules for passive targeting strategy in melanoma.
The objective of this work consists in formulating and optimizing the encapsulation of siRNA into lipid nanocapsules (LNCs) for efficient gene therapy on melanoma cells. SiRNA LNCs were prepared from DOTAP/DOPE lipoplexes, and the siRNA dose and lipid/siRNA charge ratio were assayed to improve the stability and encapsulation yield. Cryo-TEM imaging of the siRNA lipoplexes and LNC morphology revealed a specific organization of the siRNA DOTAP/DOPE lipoplexes as well as specific lipid microstructures. No cytotoxicity of the siRNA LNCs against the melanoma SK-Mel28 cell line was observed at concentrations of up to 500 ng/mL s...
