Mouse mesoderm-specific transcript inhibits adipogenic differentiation and induces trans-differentiation into hepatocyte-like cells in 3T3-L1 preadiocytes
BMC Res Notes. 2022 May 10;15(1):164. doi: 10.1186/s13104-022-06051-x.ABSTRACTOBJECTIVE: The mesoderm-specific transcript (Mest) is an imprinted gene that is transcribed from the paternal allele. It is a marker of adipose tissue expansion; however, it is uncertain whether Mest expression promotes or suppresses adipogenic differentiation. To elucidate the effects of Mest expression on adipogenic differentiation, we transfected an expression vector or siRNA for mouse Mest into 3T3-L1 mouse preadipocyte cell line.RESULTS: In differentiated 3T3-L1 adipocytes, Mest overexpression decreased lipid accumulation. Conversely, gene s...

Effect of miR-381-3p/FGF7 axis on the osteogenic differentiation of bone marrow mesenchymal stem cells through MEK/ERK signaling pathway
Tissue Cell. 2022 Apr 1;76:101791. doi: 10.1016/j.tice.2022.101791. Online ahead of print.ABSTRACTAlthough microRNAs (miRNAs) exert an important role in the osteogenesis of mesenchymal stem cells (MSCs), the effect of miR-381-3p on the osteogenic differentiation in MBD‑MSCs is still unclear. The BMMSCs from patients with MBD (MBD‑MSC) or normal participants (Normal‑MSC) were isolated and induced to differentiation with dexamethasone. BMMSCs were transfected with miR-381-3p mimic, miR-381-3p inhibitor, and FGF7 siRNA to regulate the expression of miR-381-3p or FGF7. The direct binding between miR-381-3p and FGF7 was p...

Oxygen and Steroids Affect the Regulatory Role of Natriuretic Peptide Receptor-C on Surfactant Secretion by Type II Cells
Am J Physiol Lung Cell Mol Physiol. 2021 Oct 20. doi: 10.1152/ajplung.00300.2021. Online ahead of print.ABSTRACTAtrial natriuretic peptide (ANP) and its receptors Natriuretic peptide receptor (NPR)-A and NRP-C are all highly expressed in alveolar epithelial type II cells (AEC2s) in the late gestation ovine fetal lung and are dramatically decreased postnatally. However, of all the components, NPR-C stimulation inhibits ANP-mediated surfactant secretion. Since alveolar oxygen increases dramatically after birth, and steroids are administered to mothers antenatally to enhance surfactant lung maturity, we investigated the effec...

A Primate-Specific RNA-Binding Protein (RBMXL3) Is Involved in Glucocorticoid Regulation of Human Pulmonary Surfactant Protein B (SP-B) mRNA Stability
Am J Physiol Lung Cell Mol Physiol. 2021 Mar 10. doi: 10.1152/ajplung.00022.2020. Online ahead of print.ABSTRACTThe ability of pulmonary surfactant to reduce alveolar surface tension requires adequate levels of surfactant protein B (SP-B). Dexamethasone (DEX) increases human SP-B expression, in part, through increased SP-B mRNA stability. A 30 nt-long hairpin element (RBE) in the 3'-untranslated region of human SP-B mRNA mediates both DEX-induced and intrinsic mRNA stability, but the mechanism is unknown. Proteomic analysis of RBE-interacting proteins identified a primate-specific protein; RNA binding motif X-linked-like-3...

Cellular Clocks in Hyperoxia Effects on Ca2+i Regulation in Developing Human Airway Smooth Muscle.
We report that human fASM express core clock machinery (PER1, PER2, CRY1, ARNTL/BMAL1, CLOCK) that is responsive to dexamethasone and altered by O2. Disruption of the clock via siRNA-mediated PER1 or ARNTL knockdown alters store-operated calcium entry (SOCE) and [Ca2+]i response to histamine in hyperoxia. Effects of O2 on [Ca2+]i are rescued by driving expression of clock proteins, via effects on the Ca2+ channels IP3R and Orai1. These data reveal a functional fASM clock that modulates [Ca2+]i regulation, particularly in hyperoxia. Harnessing clock biology may be a novel therapeutic consideration for neonatal airway diseas...

TRPC3 overexpression and intervention in airway smooth muscle of ovalbumin-induced hyperresponsiveness and remodeling.
Abstract Transient receptor potential canonical channel 3 (TRPC3) proteins function as non-voltage-gated Ca2+ -permeable channels and play divergent roles in many processes of pathophysiology. The purpose of this study was to determine the relationship between TRPC3 expression and airway hyperresponsiveness and remodeling in ovalbumin-induced asthmatic Kunming mice. Mice were sensitized and challenged by ovalbumin to establish asthmatic model. Hematoxylin-eosin staining, hydroxyproline assay and isometric tracheal ring force measurement were used to evaluate airway remodeling and hyperresponsiveness in asthmatic m...

Vertebrate Lonesome Kinase Regulated Extracellular Matrix Protein Phosphorylation, Cell Shape, and Adhesion in Trabecular Meshwork Cells
Glaucoma, a leading cause of irreversible blindness, is commonly associated with elevated intraocular pressure (IOP) due to impaired aqueous humor (AH) drainage through the trabecular meshwork (TM). Although dysregulated production and organization of extracellular matrix (ECM) is presumed to increase resistance to AH outflow and elevate IOP by altering TM cell contractile and adhesive properties, it is not known whether regulation of ECM protein phosphorylation via the secretory vertebrate lonesome kinase (VLK) influences TM cellular characteristics. Here, we tested this possibility. Experiments carried out in this study ...
