Retracted IQGAP1 ‑siRNA inhibits proliferation and metastasis of U251 and U373 glioma cell lines
Mol Med Rep. 2023 Nov;28(5):207. doi: 10.3892/mmr.2023.13094. Epub 2023 Sep 21.ABSTRACTFollowing the publication of this paper, it was drawn to the Editor's attention by a concerned reader that the GAPDH control western blotting data shown in Fig. 1C, and other western blotting data included in Figs. 2D and 7C and D, were strikingly similar to data appearing in different form in other articles written by different authors at different research institutes. Owing to the fact that the contentious data in the above article were already under consideration for publication, or had already been published elsewhere, prior to its s...

STAT3 suppresses the AMPK α/ULK1-dependent induction of autophagy in glioblastoma cells
In this study, we found Serine-727 and Tyrosine-705 phosphorylation of STAT3 was constitutive in GBM cell lines. Tyrosine phosphorylation of STAT3 in GBM cells suppresses autophagy, whereas knockout (KO) of STAT3 increases ULK1 gene expression, increases TSC2-AMPKα-ULK1 signalling, and increases lysosomal Cathepsin D processing, leading to the stimulation of autophagy. Rescue of STAT3-KO cells by the enforced expression of wild-type (WT) STAT3 reverses these pathways and inhibits autophagy. Conversely, expression of Y705F- and S727A-STAT3 phosphorylation deficient mutants in STAT3-KO cells did not suppress autophagy. Inhi...

Rewiring of lactate-IL-1 β auto-regulatory loop with Clock-Bmal1: A feed-forward circuit in glioma
Mol Cell Biol. 2021 Jun 14:MCB0044920. doi: 10.1128/MCB.00449-20. Online ahead of print.ABSTRACTDe-synchronized circadian rhythm in tumors is coincident with aberrant inflammation and dysregulated metabolism. As their inter-relationship in cancer etiology is largely unknown, we investigated the link between the three in glioma. Tumor metabolite lactate- mediated increase in pro-inflammatory cytokine IL-1β was concomitant with elevated levels of core circadian regulators Clock and Bmal1. siRNA mediated knockdown of Bmal1 and Clock decreased (i) LDHA and IL-1β levels and (ii) release of lactate and pro-inflammatory cytokin...

Lentivirus vector ‑mediated genetic manipulation of oncogenic pathways induces tumor formation in rabbit brain
Mol Med Rep. 2021 Jun;23(6):422. doi: 10.3892/mmr.2021.12061. Epub 2021 Apr 13.ABSTRACTTranslation of promising experimental therapies from rodent models to clinical success has been complicated as the novel therapies often fail in clinical trials. Existing rodent glioma models generally do not allow for preclinical evaluation of the efficiency of novel therapies in combination with surgical resection. Therefore, the aim of the present study was to develop a larger animal model utilizing lentivirus vector‑mediated oncogenic transformation in the rabbit brain. Lentiviruses carrying constitutively active AKT and H‑Ras on...

Silencing of ZFP36L2 increases sensitivity to temozolomide through G2/M cell cycle arrest and BAX mediated apoptosis in GBM cells.
Abstract Despite the advancements in primary brain tumour diagnoses and treatments, the mortality rate remains high, particularly in glioblastoma (GBM). Chemoresistance, predominantly in recurrent cases, results in decreased mean survival of patients with GBM. We aimed to determine the chemosensitisation and oncogenic characteristics of zinc finger protein 36-like 2 (ZFP36L2) in LN18 GBM cells via RNA interference (RNAi) delivery. We conducted a meta-analysis of microarray datasets and RNAi screening using pooled small interference RNA (siRNA) to identify the druggable genes responsive to GBM chemosensitivity. Tem...

MicroRNA ‑186‑5p downregulation inhibits osteoarthritis development by targeting MAPK1.
In conclusion, the present results indicated that miR‑186‑5p could attenuate IL‑1β‑induced chondrocyte inflammation damage by increasing MAPK1 expression, suggesting that miR‑186‑5p may be used as a potential therapeutic target for OA. PMID: 33537828 [PubMed - in process]

p53 affects epigenetic signature on SOCS1 promoter in response to TLR4 inhibition.
Abstract Suppressor of cytokine signaling (SOCS1) functions as a negative regulator of toll-like receptor (TLR) induced inflammatory signaling. As silencing of SOCS1 is concomitant with elevated TLR4 levels in glioblastoma, we investigated the effect of TLR4 inhibition on SOCS1 expression. Pharmacological inhibition of TLR4 signaling by TAK242 or its siRNA-mediated knockdown in p53 mutant or wild-type glioma cells resulted in either increased or decreased SOCS1 expression and promoter activity, respectively. Genetic manipulation of p53 indicated that SOCS1 expression upon TLR4 inhibition is dependent on p53 mutati...
