Proteomic analysis of STEAP1 knockdown in human LNCaP prostate cancer cells
Biochim Biophys Acta Mol Cell Res. 2023 Jun 12:119522. doi: 10.1016/j.bbamcr.2023.119522. Online ahead of print.ABSTRACTProstate cancer (PCa) continues to be one of the most common cancers in men worldwide. The six transmembrane epithelial antigen of the prostate 1 (STEAP1) protein is overexpressed in several types of human tumors, particularly in PCa. Our research group has demonstrated that STEAP1 overexpression is associated with PCa progression and aggressiveness. Therefore, understanding the cellular and molecular mechanisms triggered by STEAP1 overexpression will provide important insights to delineate new strategies...

HIF-1 α and Nrf2 regulates hypoxia induced overexpression of DDAH1 through promoter activation in prostate cancer
In this study, we investigated the molecular mechanism involved in the overexpression of DDAH1 in PCa and examined its potential role as a therapeutic target. We observed that DDAH1expression is elevated in PCa (PC3, LNCaP, and DU145) cell lines under hypoxia. ChIP and reporter assay results confirmed that DDAH1 expression is positively regulated by HIF-1α through directly binding to the hypoxia response elements (HRE) located within the promoter region between - 1242/- 1238 upstream of its transcription start site (TSS). Under hypoxia, HIF-1α is translocated into the nucleus and activates its target gene expression in P...

Enhanced autophagy promotes radiosensitivity by mediating Sirt1 downregulation in RM-1 prostate cancer cells
Biochem Biophys Res Commun. 2022 Apr 6;609:84-92. doi: 10.1016/j.bbrc.2022.03.142. Online ahead of print.ABSTRACTAutophagy is a double-edged sword that affects tumor progression by promoting cell survival or death depending on different living contexts. The concrete mechanism by which autophagy modulates the efficacy of radiotherapy for prostate cancer (PC) remains unclear. We exposed RM-1 PC cells to X-ray and explored the role of autophagy in radiation injury. Our results showed increased apoptosis and autophagy levels in RM-1 cells after radiation. Pharmacological inhibition of autophagy by chloroquine significantly mit...

Molecular interaction of HIC, an agonist of P2Y1 receptor, and its role in prostate cancer apoptosis
Int J Biol Macromol. 2021 Aug 20;189:142-150. doi: 10.1016/j.ijbiomac.2021.08.103. Online ahead of print.ABSTRACTProstate cancer is a heterogeneous, slow growing asymptomatic cancer that predominantly affects man. A purinergic G-protein coupled receptor, P2Y1R, is targeted for its therapeutic value since it plays a crucial role in many key molecular events of cancer progression and invasion. Our previous study demonstrated that indoline derivative, 1 ((1-(2-Hydroxy-5-nitrophenyl) (4-hydroxyphenyl) methyl)indoline-4‑carbonitrile; HIC), stimulates prostate cancer cell (PCa) growth inhibition via P2Y1R. However, the mode of...

Synthesis of KUE-siRNA conjugates for prostate cancer cell-targeted gene silencing
Chembiochem. 2021 Jul 14. doi: 10.1002/cbic.202100243. Online ahead of print.ABSTRACTThe delivery of siRNAs to selectively target cells posed a great challenge in RNAi-based cancer therapy. The lack of suitable cell-targeting methods seriously restricted the advance in delivering siRNAs to extrahepatic tissues. Based on prostate-specific membrane antigen (PSMA)-targeting ligands, we here synthesized a series of KUE-siRNA conjugates and verified their effective cell uptake and gene silencing properties in prostate cancers. The results indicated that the KUE-siRNA conjugates could selectively enter PSMA + LNCaP cells, eventu...
