Clinical analysis of bacterial infection characteristics in lymphoma patients with high-dose chemotherapy combined with autologous hematopoietic stem cell transplantation-a single-centered retrospective study
CONCLUSION: High-dose chemotherapy combined with autologous hematopoietic stem cell transplantation (HDT/AHSCT) is used to treat lymphoma. Although AHSCT has made considerable strides and become safer, HDT-AHSCT infection continues to be a leading cause of morbidity and mortality associated with transplantation.PMID:37718527 | DOI:10.2174/1386207326666230915115056

Injectable and Self-Adaptive Gel Scaffold Based on Heparin Microspheres for Adipogenesis of Human Adipose-Derived Stem Cells
Biomacromolecules. 2023 Sep 18. doi: 10.1021/acs.biomac.3c00348. Online ahead of print.ABSTRACTAn injectable and self-adaptive heparin microsphere-based cell scaffold was developed to achieve adipose regeneration. Simultaneously, the cell scaffold exhibited a dynamic architecture, self-regulated glucose levels, sustained insulin delivery, and steady viscoelastic properties for adipogenesis. The dynamic cell scaffold is cross-linked by the boronate-diol interaction among heparin-based microspheres, which have boronate and maltose groups. Because of the boronate-maltose ester bonds, the gelatinous complex would be partially ...

CRISPR/Cas9-mediated deletion of adipocyte genes associated with NAFLD alters adipocyte lipid handling and reduces steatosis in hepatocytes < em > in vitro < /em >
Am J Physiol Cell Physiol. 2023 Sep 18. doi: 10.1152/ajpcell.00291.2023. Online ahead of print.ABSTRACTObesity is a major risk factor for the development of non-alcoholic fatty liver disease (NAFLD) and the subcutaneous white adipose tissue (scWAT) is the primary lipid storage depot and regulates lipid fluxes to other organs. Our previous work identified genes up-regulated in scWAT of patients with NAFLD: SOCS3, DUSP1, and SIK1. Herein, we knocked down (KD) their expression in human adipose-derived mesenchymal stem cells (hADMSCs) using CRISPR/Cas9 technology and characterized their phenotype. We found that SOCS3, DUSP1, a...

Tert-Butyl Hydroperoxide induces Ferroptosis of Bone Mesenchymal Stem cells by repressing the Prominin2/BACH1/ROS axis
CONCLUSION: Our findings revealed a mechanism that the prominin2/BACH1/ROS axis participates in BMSCs ferroptosis and the strengthening of this axis is promising to subsist BMSCs after explantation.PMID:37721001 | DOI:10.1152/ajpcell.00224.2023

ATP promotes resident CD34 < sup > + < /sup > cell migration mainly through P2Y2-Stim1-ERK/p38 pathway
Am J Physiol Cell Physiol. 2023 Sep 18. doi: 10.1152/ajpcell.00048.2023. Online ahead of print.ABSTRACTExtracellular adenosine triphosphate (ATP) is one of the most abundant biochemical constitutes within the stem cell microenvironment and is postulated to play critical roles in cell migration. However, it is unclear whether ATP regulates the cell migration of CD34+ vascular wall-resident stem/progenitor cells (VW-SCs) and participates in angiogenesis. Therefore, the biological mechanisms of cell migration mediated by ATP was determined by in vivo subcutaneous matrigel plug assay, ex vivo aortic ring assay, in vitro transw...

Phenylethanoid glycosides derived from Cistanche deserticola promote neurological functions and the proliferation of neural stem cells for improving ischemic stroke
This study aims to investigate whether PhGs treatment improves the stroke and its potential mechanisms. Adult male C57BL/6 J mice were administrated PhGs once daily for 7 days after MCAO surgery. The neurological score, and catwalk were evaluated on Day 1, 3 and 7 after ischemic stroke. Furthermore, triphenyl-2,3,5-tetrazoliumchloride (TTC) and hematoxylin-eosin (H&E) staining were used for evaluating the infarct volume and neuronal restoration. The effects of PhGs on NSCs proliferation were investigated in vitro and in vivo. Western blot was used to detect the proteins of Wnt/β-catenin signaling pathway. This study f...

Generation of a human iPSC line CIPi003-A from a patient with focal epilepsy harboring a heterozygous mutation in DEPDC5 gene
Stem Cell Res. 2023 Aug 28;72:103195. doi: 10.1016/j.scr.2023.103195. Online ahead of print.ABSTRACTThe DEPDC5 gene (OMIM 614191) has been proven to be a frequent cause of familial and sporadic focal epilepsy. A human induced pluripotent stem cell (iPSC) line was generated from a child diagnosed with focal epilepsy, which was caused by DEDPC5 mutation. The iPSC line expresses high pluripotency markers, carries the DEDPC5 mutation, and can differentiate into three germ layers in vitro. The iPSC lines offer a promising technique for studying the pathogenesis and conducting drug screening of DEDPC5-related epilepsy.PMID:37722...

Cell-nanocarrier drug delivery system: a promising strategy for cancer therapy
Drug Deliv Transl Res. 2023 Sep 18. doi: 10.1007/s13346-023-01429-1. Online ahead of print.ABSTRACTTumor targeting has been a great challenge for drug delivery systems. A number of nanotechnology-derived drug carriers have been developed for cancer treatment to improve efficacy and biocompatibility. Among them, the emergence of cell-nanocarriers has attracted great attention, which simulates cell function and has good biocompatibility. They can also escape the clearance of reticuloendothelial system, showing a long-cycle effect. The inherent tumor migration and tumor homing ability of cells increase their significance as t...

In vitro spermatogenesis in artificial testis: current knowledge and clinical implications for male infertility
Cell Tissue Res. 2023 Sep 18. doi: 10.1007/s00441-023-03824-z. Online ahead of print.ABSTRACTMen's reproductive health exclusively depends on the appropriate maturation of certain germ cells known as sperm. Certain illnesses, such as Klinefelter syndrome, cryptorchidism, and syndrome of androgen insensitivity or absence of testis maturation in men, resulting in the loss of germ cells and the removal of essential genes on the Y chromosome, can cause non-obstructive azoospermia. According to laboratory research, preserving, proliferating, differentiating, and transplanting spermatogonial stem cells or testicular tissue could...

Chronic inflammation on gingiva-derived mesenchymal stem cells
Bioinformation. 2023 Jan 31;19(1):138-142. doi: 10.6026/97320630019138. eCollection 2023.ABSTRACTThe impact of inflammatory response on the biological characteristics of GMSCs has been the subject of studies, with conflicting findings. In order to more fully understand the effects of the localized inflammatory environment, the current study assessed the intensity and differentiating capacity of GMSCs derived from healthy periodontal tissues (H-GMSC) and GMSC derived from inflamed periodontal tissues (I-GMSC) tissues. Cells from every well were taken out and counted using a hemocytometer every three days for a total of 12 d...
