Research Progress of Metformin in the Treatment of Oral Squamous Cell Carcinoma
Endocrinology. 2023 Sep 20:bqad139. doi: 10.1210/endocr/bqad139. Online ahead of print.ABSTRACTOral squamous cell carcinoma (OSCC) is one of the most common malignancies with high mortality, posing a great threat to both human physical and mental health. With the advancement of scientific research, a variety of cancer therapies have been used for OSCC treatment. However, the prognosis of OSCC shows no significant improvement. Metformin has been recognized as the first-line drug for the treatment of diabetes, and recent studies have shown that metformin has a remarkable suppressive effect on tumor progression. Metformin can...

Metformin Ameliorates Postoperative Cognitive Dysfunction through Regulation of the AMPK/SIRT1 Pathway
CONCLUSIONS: Through the AMPK/SIRT1 pathway, metformin can boost autophagy and reduce oxidative stress in cortical microglia in older rats, in turn improving postoperative cognitive function.PMID:37735113 | DOI:10.31083/j.jin2205110

AMPK activators suppress cholesterol accumulation in macrophages via suppression of the mTOR pathway
In this study, we found that activation of AMPK by AICAR and metformin inhibits foam cell formation via suppression of mTOR in macrophages. Specifically, activation of AMPK indirectly reduced the phosphorylation level of mTOR at Ser2448 and promoted the expression of cholesterol efflux transporters and cholesterol efflux. These inhibitory effects on foam cell formation were counteracted by mTOR activators. Metformin, a more nonspecific AMPK activator than AICAR, appears to inhibit foam cell formation via anti-inflammatory effects in addition to suppression of the mTOR pathway. The results of this study suggest that the dev...

Boningmycin induces AMPK-mediated endoplasmic reticulum-associated degradation of PD-L1 protein in human cancer cells
In conclusion, it is the first report demonstrating that BON decreases PD-L1 protein level through AMPK-mediated endoplasmic reticulum-associated degradation pathway. These findings will benefit the clinical transformation of BON and aid in the elucidation of molecular mechanism of BLM-related compounds.PMID:37717372 | DOI:10.1016/j.intimp.2023.110905

SPIOMET4HEALTH —efficacy, tolerability and safety of lifestyle intervention plus a fixed dose combination of spironolactone, pioglitazone and metformin (SPIOMET) for adolescent girls and young women with polycystic ovary syndrome: study protocol for a multicentre, randomised, double-blind, placebo-controlled, fo ur-arm, parallel-group, phase II clinical trial
DiscussionThe present study will be the first to evaluate —in a randomised, double-blind, placebo-controlled way—the efficacy, tolerability and safety of SPIOMET treatment for early PCOS, on top of a lifestyle intervention.Trial registrationEudraCT 2021 –003177-58. PLGA-based microspheres loaded with metformin hydrochloride: modified double emulsion method preparation, optimization, characterization, and in vitro evaluation
Ann Pharm Fr. 2023 Sep 12:S0003-4509(23)00105-0. doi: 10.1016/j.pharma.2023.09.002. Online ahead of print.ABSTRACTThe modified solvent removal method was used to encapsulate metformin hydrochloride (MH) within poly(lactic-co-glycolic acid) (PLGA) microspheres. The study investigated the effect of varying polymer concentrations on the loading and release of the drug from the microspheres. The encapsulation process involved using a double emulsion method, resulting in microspheres with particle diameters ranging from approximately 4.4 µm to 2.7 µm. The study achieved high encapsulation efficiencies, ranging from 81% to 90%...
