Remote Activation of Enzyme Nanohybrids for Cancer Prodrug Therapy Controlled by Magnetic Heating
ACS Nano. 2023 Jun 26. doi: 10.1021/acsnano.3c01599. Online ahead of print.ABSTRACTHerein, we have developed nanohybrids (nHs) to remotely activate a therapeutic enzyme for its use in Directed Enzyme Prodrug Therapy (DEPT). The coencapsulation of magnetic nanoparticles (MNPs) with horseradish peroxidase (HRP) using biomimetic silica as an entrapment matrix was optimized to obtain nanosized hybrids (∼150 nm) for remote activation of the therapeutic enzyme. HRP converts indole-3-acetic acid (3IAA) into peroxylated radicals, whereas MNPs respond to alternating magnetic fields (AMFs) becoming local hotspots. The AMF applicat...

Multifunctional Low-Generation Dendrimer Nanogels as an Emerging Probe for Tumor-Specific CT/MR Dual-Modal Imaging
Biomacromolecules. 2023 Jan 6. doi: 10.1021/acs.biomac.2c01403. Online ahead of print.ABSTRACTThe development of nanoprobes that have amplified enhanced permeability and retention (EPR) effect is crucial for their precise cancer diagnosis performance. Here, we present the development of functional dendrimer-based nanogels (DNGs) with the generation three primary amine-terminated poly(amidoamine) (PAMAM) dendrimers (G3·NH2) cross-linked by N,N'-bis(acryloyl) cystamine (BAC). The DNGs were prepared through a Michael addition reaction between G3·NH2 dendrimers and BAC via an inverse microemulsion method and entrapped with g...

Acetic Acid Mediated for One-Pot Synthesis of Novel Pyrazolyl < em > s < /em > -Triazine Derivatives for the Targeted Therapy of Triple-Negative Breast Tumor Cells (MDA-MB-231) < em > via < /em > EGFR/PI3K/AKT/mTOR Signaling Cascades
Pharmaceutics. 2022 Jul 27;14(8):1558. doi: 10.3390/pharmaceutics14081558.ABSTRACTHere, we described the synthesis of novel pyrazole-s-triazine derivatives via an easy one-pot procedure for the reaction of β-dicarbonyl compounds (ethylacetoacetate, 5,5-dimethyl-1,3-cyclohexadione or 1,3-cyclohexadionone) with N,N-dimethylformamide dimethylacetal, followed by addition of 2-hydrazinyl-4,6-disubstituted-s-triazine either in ethanol-acetic acid or neat acetic acid to afford a novel pyrazole and pyrazole-fused cycloalkanone systems. The synthetic protocol proved to be efficient, with a shorter reaction time and high chemical y...

The Effect of Resveratrol on the Composition and State of Lipids and the Activity of Phospholipase A2 During the Excitation and Regeneration of Somatic Nerves
Conclusion It was established that both the nerve excitation conduction and the transection of somatic nerves lead to profound changes in the lipid composition. Not only the number of phospholipids, but also the phospholipid fatty acid composition is changing. The key factors responsible for the detected changes are phospholipase A2 and phospholipase C activities. In the first place of the nerve fiber degenerative processes were both Ca 2+-dependent and Ca 2+-independent phospholipase A2 activation in the fiber’s distal part, which associated with of the axonal transport blocking. Another factor affecting the nerv...

Bioactivities and Extract Dereplication of Actinomycetales Isolated From Marine Sponges
This study aimed to contribute to the need of finding new and more effective bioactive molecules against several of the earlier mentioned threats faced by human kind nowadays. For this, eighteen species from the Actinomycetales previously isolated from Erylus spp. sponges collected in Portuguese marine waters (Açores, Madeira, and continental shelf) were screened for antimicrobial, anti-cancer, anti-parasitic and anti-obesogenic activities. Materials and Methods Biological Material The bacteria under study belong to the order Actinomycetales within the phylum Actinobacteria and were isolated from marine sponges o...
