Emerging Role of Ferroptosis in Breast Cancer: Characteristics, Therapy, and Translational Implications for the Present and Future
We present a fundamental analysis of the actual understanding of molecular mechanisms along with regulatory networks associated with ferroptosis, expected physiological functions in growth concealment, ferroptosis-associated differentially expressed genes, treatment targeting potential, and recent advances in the development of therapeutic strategies in this review.PMID:37711099 | DOI:10.2174/1566524023666230913105735

Ferroptosis and EMT: key targets for combating cancer progression and therapy resistance
Cell Mol Life Sci. 2023 Aug 19;80(9):263. doi: 10.1007/s00018-023-04907-4.ABSTRACTIron-dependent lipid peroxidation causes ferroptosis, a form of regulated cell death. Crucial steps in the formation of ferroptosis include the accumulation of ferrous ions (Fe2+) and lipid peroxidation, of which are controlled by glutathione peroxidase 4 (GPX4). Its crucial role in stopping the spread of cancer has been shown by numerous studies undertaken in the last ten years. Epithelial-mesenchymal transition (EMT) is the process by which epithelial cells acquire mesenchymal characteristics. EMT is connected to carcinogenesis, invasivenes...

Black phosphorus quantum dots induced ferroptosis in lung cell via increasing lipid peroxidation and iron accumulation
Food Chem Toxicol. 2023 Jul 20:113952. doi: 10.1016/j.fct.2023.113952. Online ahead of print.ABSTRACTBlack Phosphorus Quantum Dots (BP-QDs) have potential applications in biomedicine. BP-QDs may enter the body through the respiratory tract during grinding and crushing production and processing, causing respiratory toxicity. Ferroptosis is an oxidative, iron-dependent form of cell death. Here, respiratory toxicity of BP-QDs has been validated in mice and human bronchial epithelial cells. After 24 h of exposure to different doses (4-32 μg/mL) of BP-QDs, intracellular lipid peroxidation and iron overload occurred in Beas-2B ...

Fight Aging! Newsletter, July 3rd 2023
In this study, cultured adipose-derived stem cells (ASCs) were derived from subcutaneous white adipose tissue isolated from mice fed a normal diet. We performed senescence-associated-β-galactosidase (SA-β-gal) staining, real-time PCR, and Western blot to evaluate the levels related to cellular senescence markers. The mRNA expression levels of senescence markers were significantly increased in the later passages of ASCs. We show that light activation reduced the expression of senescent genes, and SA-β-Gal in all cells at passages. Moreover, the light-activated ASCs-derived exosomes decrease the expression of senes...

CISD2 promotes lung squamous carcinoma cell migration and invasion via the TGF- β1-induced Smad2/3 signaling pathway
CONCLUSION: In conclusion, our research illustrates that CISD2 is highly expressed in LUSC and may facilitate LUSC proliferation and metastasis. Thus, CISD2 may serve as an independent prognostic marker and possible treatment target for LUSC.PMID:37249759 | DOI:10.1007/s12094-023-03222-5

SIRT3-mediated autophagy contributes to ferroptosis-induced anticancer by inducing the formation of BECN1-SLC7A11 complex
Biochem Pharmacol. 2023 May 15:115592. doi: 10.1016/j.bcp.2023.115592. Online ahead of print.ABSTRACTFerroptosis is an autophagy-dependent cell death associated with iron accumulation and lipid peroxidation, which plays a crucial part in anticancer activity. Sirtuin 3 (SIRT3) positively regulates autophagy by phosphorylation of activated protein kinase (AMPK). However, whether SIRT3-mediated autophagy can inhibit the cystine/glutamate antiporter (system Xc-) activity by inducing the formation of a BECN1-SLC7A11 complex and consequently promote induction of ferroptosis is unknown. Using both in vitro and in vivo models, we ...

TIGAR protects against adenine-induced ferroptosis in human proximal tubular epithelial cells by activating the mTOR/S6KP70 axis
Nutr Cancer. 2023 May 4:1-9. doi: 10.1080/01635581.2023.2203353. Online ahead of print.ABSTRACTTP53-induced glycolysis and apoptosis regulator (TIGAR) acts as a switch for nephropathy, but its underlying mechanism is still unclear. The purpose of this study was to explore the potential biological significance and underlying mechanism of TIGAR in modulating adenine-induced ferroptosis in human proximal tubular epithelial (HK-2) cells. HK-2 cells under- or overexpressing TIGAR were challenged with adenine to induce ferroptosis. The levels of reactive oxygen species (ROS), iron, malondialdehyde (MDA), and glutathione (GSH) we...
