The role of LOXL2 induced by glucose metabolism-activated NF- κB in maintaining drug resistance through EMT and cancer stemness in gemcitabine-resistant PDAC
J Mol Med (Berl). 2023 Sep 22. doi: 10.1007/s00109-023-02369-6. Online ahead of print.ABSTRACTGemcitabine is considered a standard treatment for pancreatic cancer, but developing drug resistance greatly limits the effectiveness of chemotherapy and increases the rate of recurrence. Lysyl oxide-like 2 (LOXL2) is highly expressed in pancreatic cancer and is involved in carcinogenesis and EMT regulation. However, studies on the role of LOXL2 in drug resistance are limited. Here, we investigated the mechanism of LOXL2 induction and the effect of LOXL2 on EMT and CSC in gemcitabine-resistant pancreatic cancer. Glucose metabolism...

Interaction between N < sup > 6 < /sup > -methyladenosine modification and the tumor microenvironment in colorectal cancer
Mol Med. 2023 Sep 22;29(1):129. doi: 10.1186/s10020-023-00726-2.ABSTRACTThe incidence and mortality of colorectal cancer (CRC) are rapidly increasing worldwide. Recently, there has been significant attention given to N6-methyladenosine (m6A), the most common mRNA modification, especially for its effects on CRC development. It is important to note that the progression of CRC would be greatly hindered without the tumor microenvironment (TME). The interaction between CRC cells and their surroundings can activate and influence complex signaling mechanisms of epigenetic changes to affect the survival of tumor cells with a malig...

The prognostic significance of a negative PSMA-PET scan prior to salvage radiotherapy following radical prostatectomy
CONCLUSION: Favorable bRFS after SRT in patients with BR and negative PSMA-PET following RP was achieved. These data support the usage of early SRT for patients with negative PSMA-PET findings.PMID:37736808 | DOI:10.1007/s00259-023-06438-3

Evaluation of enhanced permeability effect and different linear energy transfer of radionuclides in a prostate cancer xenograft model
In conclusion, we found that specific targeting might negatively influence normal organ uptake when targeting secreted antigens. Furthermore, different energy deposition i.e. linear energy transfer of a radionuclide might have diverse effects on receptor expression and cell proliferation in tumors.PMID:37736493 | PMC:PMC10509292

PARylated PDHE1 α generates acetyl-CoA for local chromatin acetylation and DNA damage repair
Nat Struct Mol Biol. 2023 Sep 21. doi: 10.1038/s41594-023-01107-3. Online ahead of print.ABSTRACTChromatin relaxation is a prerequisite for the DNA repair machinery to access double-strand breaks (DSBs). Local histones around the DSBs then undergo prompt changes in acetylation status, but how the large demands of acetyl-CoA are met is unclear. Here, we report that pyruvate dehydrogenase 1α (PDHE1α) catalyzes pyruvate metabolism to rapidly provide acetyl-CoA in response to DNA damage. We show that PDHE1α is quickly recruited to chromatin in a polyADP-ribosylation-dependent manner, which drives acetyl-CoA generation to su...

Epigenetic reprogramming of a distal developmental enhancer cluster drives SOX2 overexpression in breast and lung adenocarcinoma
Nucleic Acids Res. 2023 Sep 22:gkad734. doi: 10.1093/nar/gkad734. Online ahead of print.ABSTRACTEnhancer reprogramming has been proposed as a key source of transcriptional dysregulation during tumorigenesis, but the molecular mechanisms underlying this process remain unclear. Here, we identify an enhancer cluster required for normal development that is aberrantly activated in breast and lung adenocarcinoma. Deletion of the SRR124-134 cluster disrupts expression of the SOX2 oncogene, dysregulates genome-wide transcription and chromatin accessibility and reduces the ability of cancer cells to form colonies in vitro. Analysis...

Fructose-induced mTORC1 Activation Promotes Pancreatic Cancer Progression through Inhibition of Autophagy
In this study, we investigated the functional association between fructose utilization and pancreatic ductal adenocarcinoma (PDAC) progression. Fructose could be taken up and metabolized by PDAC cells and provided an adaptive survival mechanism for PDAC cells under glucose deficient conditions. GLUT5-mediated fructose metabolism maintained the survival, proliferation, and invasion capacities of PDAC cells in vivo and in vitro. Fructose metabolism not only provided ATP and biomass to PDAC cells but also conferred metabolic plasticity to the cells, making them more adaptable to the tumor microenvironment. Mechanistically, fr...

Single-cell and bulk sequencing analyses reveal the immune suppressive role of PTPN6 in glioblastoma
Aging (Albany NY). 2023 Sep 21;15. doi: 10.18632/aging.205052. Online ahead of print.ABSTRACTGlioblastoma (GBM) is a highly malignant brain cancer with a poor prognosis despite standard treatments. This investigation aimed to explore the feasibility of PTPN6 to combat GBM with immunotherapy. Our study employed a comprehensive analysis of publicly available datasets and functional experiments to assess PTPN6 gene expression, prognostic value, and related immune characteristics in glioma. We evaluated the influence of PTPN6 expression on CD8+ T cell exhaustion, immune suppression, and tumor growth in human GBM samples and mo...
