NSAIDs do not prevent exercise-induced performance deficits or alleviate muscle soreness: a placebo-controlled randomized, double-blinded, cross-over study
Non-steroidal anti-inflammatory drugs (NSAIDs) are frequently consumed by athletes to manage muscle soreness, expedite recovery, or improve performance. Despite the prevalence of NSAID use, their effects on muscle soreness and performance, particularly when administered prophylactically, remain unclear. This randomized, double-blind, counter-balanced, crossover study examined the effect of consuming a single dose of each of three NSAIDs (celecoxib, 200  mg; ibuprofen, 800 mg; flurbiprofen, 100 mg) or placebo 2 h before on muscle soreness and performance following an acute plyometric training session. Emerging drugs for the treatment of irritability associated with autism spectrum disorder
Expert Opin Emerg Drugs. 2024 Jan 31:1-12. doi: 10.1080/14728214.2024.2313650. Online ahead of print.ABSTRACTINTRODUCTION: Autism spectrum disorder (ASD) is an early-onset disorder with a prevalence of 1% among children and reported disability-adjusted life years of 4.31 million. Irritability is a challenging behavior associated with ASD, for which medication development has lagged. More specifically, pharmacotherapy effectiveness may be limited against high adverse effects (considering side effect profiles and patient medication sensitivity); thus, the possible benefits of pharmacological interventions must be balanced ag...

Synthesis of Chalcone Derivatives as COX-2 Inhibitory Activity for Ischemic Stroke Treatment
Cyclooxygenase (COX)-2 is a crucial mediator contributing to blood –brain barrier damage during cerebral ischemia. The ability of target compounds to inhibit COX-1 and COX-2 enzymes was determined. Compound4a showed a high COX-2 inhibitory potency and selectivity, which was higher activity than observed for the positive control celecoxib. A docking study was performed for compound4a to explain its interaction with the COX-2 receptor active site. (Source: Chemistry of Natural Compounds)

Synthesis and biological evaluation of new nicotinic acid derivatives as potential anti-inflammatory agents with enhanced gastric safety profile
Bioorg Chem. 2024 Mar;144:107136. doi: 10.1016/j.bioorg.2024.107136. Epub 2024 Jan 19.ABSTRACTTwo innovative series derived from nicotinic acid scaffold were synthesized and evaluated for their anti-inflammatory activity. Ibuprofen, celecoxib and indomethacin were used as standard drugs. All the newly synthesized compounds were in vitro screened for their anti-inflammatory activity adopting 3-(4,5-dimethylthiazol-2-yl)2,5-diphenyltetrazolium bromide dye (MTT), as well as Griess assays. The results showed that all compounds exhibited significant anti-inflammatory activity without affecting the viability of the macrophages c...

Hybrids of selective COX-2 inhibitors and active derivatives of edaravone as COX-2 selective NSAIDs with free radical scavenging activity: Design, synthesis and biological activities
In conclusion, this study provided a novel class of coxibs containing edaravone moiety as COX-2 selective NSAIDs with free radical scavenging activity and the candidate compound WYZ90 showed not only similar selective COX-2 inhibitory and analgesic activity to celecoxib but also better free radical scavenging and antioxidant activity than edaravone.PMID:38266553 | DOI:10.1016/j.ejmech.2024.116155 (Source: European Journal of Medicinal Chemistry)

Evaluation of the antimicrobial efficacy of different nonsteroidal anti-inflammatory drugs alone or in combination with calcium hydroxide intracanal medicament: an in vitro study
This study explored the antimicrobial effects of ketoprofen, piroxicam, and celecoxib alone or combined with calcium hydroxide (CH) against two strains ofEnterococcus faecalis (E. faecalis) and assessed the influence of such combinations on the pH of CH. Minimum inhibitory concentrations (MICs) of the three tested NSAIDs were determined. Tested pastes were placed into wells punched in seeded agar plates and the bacterial inhibition zones were measured. Antibiofilm activity was assessed against 3 weeks of biofilm induced in bovine dentine blocks. The pH of the pastes was measured at four-time intervals. MIC values were 3.12...

Revisiting prostaglandin E2: A promising therapeutic target for osteoarthritis
Clin Immunol. 2024 Mar;260:109904. doi: 10.1016/j.clim.2024.109904. Epub 2024 Jan 21.ABSTRACTOsteoarthritis (OA) is a complex disease characterized by cartilage degeneration and persistent pain. Prostaglandin E2 (PGE2) plays a significant role in OA inflammation and pain. Recent studies have revealed the significant role of PGE2-mediated skeletal interoception in the progression of OA, providing new insights into the pathogenesis and treatment of OA. This aspect also deserves special attention in this review. Additionally, PGE2 is directly involved in pathologic processes including aberrant subchondral bone remodeling, car...
