Cancer Biol Ther. 2021 Oct 25:1-5. doi: 10.1080/15384047.2021.1991739. Online ahead of print.ABSTRACTMetastatic pancreatic adenocarcinoma (PDAC) is a rapidly lethal disease, with less than half of patients surviving 12 months, and 5-year survival approximately 3%. These outcomes are in large part due to a lack of effective medical and surgical therapies for metastatic PDAC. Herein, we present the case of a patient with oligometastatic liver recurrence of BRCA2-mutated PDAC following a curative-intent resection. Through a combination of systemic chemotherapy, metastasectomy, radiotherapy, and subsequent targeted therapy wit...

Conclusion: Modulating B cell development, neoangiogenesis and fibrosis, and SDF-1/CXCL12 may play a role in IgG4-RD. The higher levels observed in IgG4-RD, as compared to SSj, which closely mimics the disease, can be related to a different pattern of lesions, with prevalent fibrosis seen in IgG4-RD. Taken together, these findings suggest that drugs acting on the CXCL12/CXCR4/CXCR7 axis may affect IgG4-RD.

Experimental &Molecular Medicine, Published online: 26 October 2021; doi:10.1038/s12276-021-00687-8Boosting the activity of a tumor suppressor pathway in pancreatic cancer may reduce the risk of metastasis. Reduced expression of a tumor suppressor gene called Krüppel-like factor 10 (KLF10) is found in breast, lung and pancreatic cancers. KLF10 deficiency in pancreatic cancer is linked to accelerated cancer progression and metastasis, but the underlying mechanisms are unclear. In experiments on mouse models and human cell cultures, Hui-Ju Ch’ang at the National Institute of Cancer Research in Zhunan, Taiwan, ...

In this study, chemopreventive effects of O-Hexadecyl- γ-glutamyl-S-benzyl-cysteinyl-D-phenyl glycine-Ethylester (HGBPE), which we previously synthesized to inhibit GSTP1 activity, was analyzed for its effect on the prevention of a rat pancreatic carcinogenesis model induced by 7,12-dimethyl-benzanthracene (DMBA). Rats administered with DMBA were group ed into five cohorts. In the treated group I, which was treated neither with HGBPE nor vehicle, sequential appearance of precancerous lesions, ductal complexes, and adenocarcinoma was confirmed as previously reported. We also confirmed in this group that mutations of K...

iane J. Bruns Felix C. Popp In pancreatic ductal adenocarcinoma (PDAC), the tumor stroma constitutes most of the cell mass and contributes to therapy resistance and progression. Here we show a hitherto unknown metabolic cooperation between pancreatic stellate cells (PSCs) and tumor cells through Interleukin 17B/Interleukin 17B receptor (IL-17B/IL-17RB) signaling. Tumor-derived IL-17B carrying extracellular vesicles (EVs) activated stromal PSCs and induced the expression of IL-17RB. PSCs increased oxidative phosphorylation while reducing mitochondrial turnover. PSCs activated tumor cells in a feedback loop. Tumor cell...

In conclusion, TPS levels increase late in PDAC progression and hold no potential as a biomarker for early detection.

Neoadjuvant chemotherapy (NAC) is being utilized more frequently in patients with pancreatic adenocarcinoma (PDAC). Not all patients can tolerate modern multiagent regimens due to toxicity profiles or pre-existing comorbidities. We utilized institutional data and the National Cancer Database (NCDB) to investigate if the time from diagnosis to NAC start is associated with survival from surgery for PDAC.

Pancreatic adenocarcinoma is the fourth leading cause cancer-related death. Previous studies have recognized that a high proportion of patients with localized cancer were not undergoing surgical resection. Our study aimed to examine the factors associated with pancreatic surgical intervention to identify opportunities for targeted improvement.

Pancreatic ductal adenocarcinoma (PDAC) is associated with increased venous thromboembolism (VTE). We  sought to compare rates of VTE and bleeding complications in patients receiving extended postoperative thromboprophylaxis (EPT) to those who did not, and to identify factors associated with increased VTE risk after pancreatic resection for PDAC.
