Impairment of the Gnα11-controlled expression of claudin-1 and MMP-9 and collective migration of human breast cancer MCF-7 cells by DHEAS

We address here DHEAS effects on the estrogen receptor-positive metastatic human breast cancer cell line MCF-7. We focus on DHEAS-mediated signaling that might influence expression of claudin-1 and matrix metalloproteinase-9 (MMP-9), both known to be critical factors for migration and invasiveness of various cancers, including breast cancer cells.Physiological concentrations of DHEAS trigger persistent phosphorylation of Erk1/2 in MCF-7 cells. Exposure of these cells for 24 h to 1 μM DHEAS also leads to a significant reduction of claudin-1 expression that cannot be prevented by high concentrations of the steroid sulfatase inhibitor STX64, indicating that desulfation and further conversion of DHEAS to some other steroid hormone is not required for this action. The UK FAST trial tested 5 fractions (Fr) of 5.7 Gy and 6.0 Gy against 25 Fr of 2.0 Gy in women prescribed whole breast radiotherapy (no boost) after local excision of early breast cancer. Analysis of primary endpoint (normal tissue effects [by photograph]) showed that the 28.5 Gy/5 Fr regimen appeared similar to control. Further follow-up now enables analysis of 10-year outcomes.

To the Editor: We thank Professor Spratt for his editorial, which accompanied publication of our manuscript that assessed Ki67 as a prognostic and predictive biomarker after radiation therapy in localized prostate cancer (1, 2). The editorial mentions a rather small increase in biochemical recurrence per unit increase in Ki67, which warrants clarification. We assessed Ki67 as a continuous variable using a percentage scale, as recommended in both the Transparent Reporting of a Multivariate Prediction Model for Individual Prognosis or Diagnosis statement (3) and recent literature relating to optimal use of Ki67 in breast cancer (4).

AbstractBreast cancer is the most common cancer in women in our country and it is usually diagnosed in the early and potentially curable stages. Nevertheless, around 20 –30% of patients will relapse despite appropriate locoregional and systemic therapies. A better knowledge of this disease is improving our ability to select the most appropriate therapy for each patient with a recent diagnosis of an early stage breast cancer, minimizing unnecessary toxicities and improving long-term efficacy.
