Patients with breast cancer (BC) and those at high risk for BC are often reluctant to initiate Tamoxifen (Tam) due to potential adverse events (AEs) such as deep vein thrombosis (DVT), pulmonary embolus (PE), acute ischemic stroke (AIS), myocardial infarction (MI) and endometrial cancer (EC). Increased risk of arterial thromboembolism in older men with breast cancer
ConclusionsWe provide population-based evidence that male breast cancer patients have a substantially elevated risk of ATE in the first year following a cancer diagnosis compared with matched controls. Care providers should consider this heightened risk when evaluating cardiovascular health in men with a recent breast cancer diagnosis.

Problem-patients in HRT
Since the Women's Health Initiative (WHI) has been stopped 2002 due to increased risk of venous thromboembolism, myocardial infarction, stroke and breast cancer, safety issues have got great concern using hormone replacement therapy (HRT). However, using lower dosages, other hormonal components and/or transdermal instead of oral application can reduce those risks although not demonstrated in a placebo-controlled study comparable to WHI. However, this has been shown in large case/control- and cohort-studies reflecting more practical conditions in contrast to WHI.

Drug interactions and pharmacogenetic factors contribute to variation in apixaban concentration in atrial fibrillation patients in routine care
In this study, we assessed the impact of interacting medication and pharmacogenetic variation to better explain apixaban concentration differences among 358 Caucasian AF patients. Genotyping (ABCG2,ABCB1,CYP3A4*22,CYP3A5*3) was performed by TaqMan assays, and apixaban quantified by mass spectrometry. Risk of Cardiovascular Diseases Among Older Breast Cancer Survivors in the United States: A Matched Cohort Study.
CONCLUSIONS: Women with a history of breast cancer were at increased risk of several CVDs, persisting into survivorship.
