Multidimensional Analysis of a Cell-Free DNA Whole Methylome Sequencing Assay for Early Detection of Gastric Cancer: Protocol for an Observational Case-Control Study
CONCLUSIONS: This is the first registered case-control study designed to investigate a stacked ensemble model integrating several cfDNA features generated from a bisulfite-free whole methylome sequencing assay. These features include methylation patterns, fragmentation profiles, and chromosomal copy number changes, with the aim of identifying the GC population. This study will determine whether multidimensional analysis of cfDNA will prove to be an effective strategy for distinguishing patients with GC from nonmalignant individuals within the Chinese population. We anticipate the THEMIS model will complement the standard-o...

Nontraditional Approaches To Enable High-Energy and Long-Life Lithium-Sulfur Batteries
Acc Chem Res. 2023 Sep 20. doi: 10.1021/acs.accounts.3c00400. Online ahead of print.ABSTRACTConspectusLithium-sulfur (Li-S) batteries are promising for automotive applications due to their high theoretical energy density (2600 Wh/kg). In addition, the natural abundance of sulfur could mitigate the global raw material supply chain challenge of commercial lithium-ion batteries that use critical elements, such as nickel and cobalt. However, due to persistent polysulfide shuttling and uncontrolled lithium dendrite growth, Li-S batteries using nonencapsulated sulfur cathodes and conventional ether-based electrolytes suffer from...

Synthesis of Peptidyl-tRNA Mimics for Structural Biology Applications
Acc Chem Res. 2023 Sep 20. doi: 10.1021/acs.accounts.3c00412. Online ahead of print.ABSTRACTConspectusProtein biosynthesis is a central process in all living cells that is catalyzed by a complex molecular machine─the ribosome. This process is termed translation because the language of nucleotides in mRNAs is translated into the language of amino acids in proteins. Transfer RNA (tRNA) molecules charged with amino acids serve as adaptors and recognize codons of mRNA in the decoding center while simultaneously the individual amino acids are assembled into a peptide chain in the peptidyl transferase center (PTC). As the nasc...

The inherited KRAS-variant as a biomarker of Cetuximab Response in NSCLC
CONCLUSIONS/DISCUSSION: Although an overall survival advantage was not achieved in KRAS-variant patients, there is potential impact of cetuximab for this genetic subset of patients. In contrast, cetuximab seems to harm non-variant patients. These findings further support the importance of genetic patient selection in trials studying the addition of systemic agents to radiotherapy.PMID:37728512 | DOI:10.1158/2767-9764.CRC-23-0084

Metastatic competency and tumor spheroid formation are independent cell states governed by RB in lung adenocarcinoma
Cancer Res Commun. 2023 Sep 20. doi: 10.1158/2767-9764.CRC-23-0172. Online ahead of print.ABSTRACTInactivation of the retinoblastoma (RB) tumor suppressor in lung adenocarcinoma is associated with the rapid acquisition of metastatic ability and the loss of lung cell lineage commitment. We previously showed that restoration of RB in advanced lung adenocarcinomas in the mouse was correlated with a decreased frequency of lineage de-committed tumors and overt metastases. To identify a causal relationship for RB and its role in reprogramming lineage commitment and reducing metastatic competency in lung adenocarcinoma, we develo...

EDIL3 as an angiogenic target of immune exclusion following checkpoint blockade
Cancer Immunol Res. 2023 Sep 20. doi: 10.1158/2326-6066.CIR-23-0171. Online ahead of print.ABSTRACTImmune checkpoint blockade (ICB) has become the standard of care for several solid tumors. Multiple combinatorial approaches have been studied to improve therapeutic efficacy. The combination of anti-angiogenesis agents and ICB has demonstrated efficacy in several cancers. To improve the mechanistic understanding of synergies with these treatment modalities, we performed serologic screens of human protein array using sera from long-term responding patients treated with ipilimumab and bevacizumab. We discovered a high-titer an...

Investigation of conditioned medium properties obtained from human umbilical cord mesenchymal stem/stromal cells preconditioned with dimethyloxalylglycine in a correlation with ultrastructural changes
Microsc Res Tech. 2023 Sep 20. doi: 10.1002/jemt.24420. Online ahead of print.ABSTRACTMesenchymal stem/stromal cells (MSCs) hold significant therapeutic value due to their regeneration abilities, migration capacity, and immunosuppressive and immunomodulatory properties. These cells secrete soluble and insoluble factors, and this complex secretome contributes to their therapeutic effect. Furthermore, stimulation of cells by various external stimuli lead to secretome modifications that can increase the therapeutic efficacy. So, this study examined the effect of dimethyloxalylglycine (DMOG), a hypoxia-mimetic agent, on secret...
