Metformin protects against retinal ischemia/reperfusion injury through AMPK-mediated mitochondrial fusion
This study aims to evaluate the protective effect of metformin against retinal I/R injury and its underlying mechanism. I/R induced reduction in retina thickness and cell number in ganglion cell layer, and metformin alleviated I/R-induced retinal injury. Both retinal I/R and simulated ischemia/reperfusion (SIR) in R28 cells down-regulated expression of mitochondrial fusion protein Mfn2 and OPA1, which led to mitochondrial fission. Metformin also alleviated damage in R28 cells, and reversed the alteration in Mfn2 and OPA1, mitochondrial fission and mitochondrial membrane potential (MMP) disruption-induced by I/R or SIR as w...

Astragaloside IV attenuates podocyte apoptosis through ameliorating mitochondrial dysfunction by up-regulated Nrf2-ARE/TFAM signaling in diabetic kidney disease
In this study, using molecular docking we found that Astragaloside IV (AS-IV), an active ingredient from traditional formula of Huangqi decoction (HQD), exerted a higher potential to promote Nrf2 escape from Keap1-Nrf2 interaction via competitively bind to amino acid sites in Keap1. When podocyte exposed to high glucose (HG) stimulation, mitochondrial morphological alterations and podocyte apoptosis were presented and accompanied by Nrf2 and mitochondrial transcription factor A (TFAM) downregulation. Mechanistically, HG promoted a decrease in mitochondria-specific electron transport chain (ETC) complexes, ATP synthesis and...

Caveolin-1 alleviates acetaminophen-induced vascular oxidative stress and inflammation in non-alcoholic fatty liver disease
In conclusion, our findings confirmed that CAV1 exerts a protective effect against vascular injury by inhibiting oxidative stress and inflammation through the PKC/MAPK pathway. Therefore, restoration of CAV1 may have clinical benefits in reducing APAP-induced vascular damage in NAFLD patients.PMID:36596386 | DOI:10.1016/j.freeradbiomed.2022.12.095

Silencing SIRT5 induces the senescence of UCB-MSCs exposed to TNF- α by reduction of fatty acid β-oxidation and anti-oxidation
Free Radic Biol Med. 2022 Sep 9:S0891-5849(22)00583-4. doi: 10.1016/j.freeradbiomed.2022.09.002. Online ahead of print.ABSTRACTTumor necrosis factor-α (TNF-α) is an inflammatory cytokine involved in cell survival, apoptosis, and homeostasis. However, the regulatory effect of TNF-α on mesenchymal stem cell (MSC) redox regulation remains unknown. The process of delaying the senescence of MSCs and maintaining antioxidation mechanism is important in transplantation therapy to treat inflammatory diseases that result from restricted immunomodulatory effects of senescent MSCs. Thus, we examined the role of TNF-α-mediated sign...

HIF-1 α Mediates Arsenic-Induced Metabolic Reprogramming in Lung Bronchial Epithelial Cells
Biol Trace Elem Res. 2022 Jun 17. doi: 10.1007/s12011-022-03315-2. Online ahead of print.ABSTRACTArsenic is a common environmental pollutant that can cause damage to multiple systems and organs in the body. The lungs are particularly sensitive to arsenic exposure, and respiratory disease is thought to be the leading cause of death from arsenic poisoning. Our previous study found that human bronchial epithelial (HBE) cells treated with NaAsO2 exhibited mitochondrial dysfunction accompanied by elevated HIF-1α; however, the molecular mechanism was unclear. The aim of the current study was to confirm the role of HIF-1α in ar...

MAPK15-ULK1 signaling regulates mitophagy of airway epithelial cell in chronic obstructive pulmonary disease
This study investigated MAPK15-ULK1 signaling in airway epithelial cells of COPD patients and its activation by cigarette smoke extract (CSE) in isolated human airway epithelial cells. Significant increased phosphorylation of MAPK15 and ULK1 (Ser555) was detected in the airway epithelium of COPD patients. This pathology was maintained in isolated primary COPD-epithelial cells. Compared to control cells, the protein expression of Beclin1 and the ratio of LC3II to LC3I were both significantly increased in COPD-epithelial cells. In human airway epithelial cells, CSE significantly increased the phosphorylation of MAPK15, ULK1 ...
