Abstract Vitiligo is an acquired condition characterized by depigmented, cutaneous lesions that result from the death of pigment‐producing cells, melanocytes. The occurrence of oxidative stress has been proposed as a pathogenetic mechanism for melanocyte degeneration in vitiligo. Therefore, in this study, we investigated the cytoprotective effects of afzelin against oxidative stress and its mechanism of action in human epidermal melanocytes. 512 ATM significantly protects against oxidative injury in human melanocyte associated with activation of Nrf2 pathway
Vitiligo is a common depigmentation disorder characterized by a loss of functional melanocytes from human epidermis, our previous study showed the increased sensitivity to H2O2-induced oxidative insults of vitiligo melanocytes is due to the dysfunction inNrf2-ARE signaling pathway. Here, we sought to determine the underlying mechanism for the aberrant expression of Nrf2-ARE signaling pathway. We found that transfection with ATM-siRNA leads to a significantly increased cell deathcaused by H2O2 and higher intracellular ROS and MDA levels in melanocytes, but have no effect on melanin synthesis and tyrosinase activity.

miR-196a-2 rs11614913 polymorphism is associated with vitiligo by affecting heterodimeric molecular complexes of Tyr and Tyrp1
In conclusion, our findings suggest that miR-196a-2 polymorphisms can regulate the Tyr levels, which influences the susceptibility of vitiligo.

Protease-activated receptor-2 activates NQO-1 via Nrf2 stabilization in keratinocytes
Conclusion: Collectively, PAR-2 activation enhanced nuclear Nrf2 translocation, and PAR-2-mediated Nrf2 activation was attributable to existing Nrf2 stabilization rather than de novo production. Our findings suggest that in addition to induction of inflammation, PAR-2 can play a chemopreventative role via Nrf2 stabilization in keratinocytes.
