MiR-125b-5p up-regulation in asthma and COPD overlap patients mediates oxidative stress and apoptosis via targeting IL6R/NLRC5/TRIAP1
Conclusions: MiR-125b-5p is up-regulated in ACO patients, and may cause oxidative stress and cell apoptosis through targeting IL6R, NLRC5, TRIAP1 genes.

IL-11 increases levels and activates circulating fibrocytes in different pulmonary hypertension animal models.
In conclusion, IL-11 promotes an increase of circulating fibrocytes, their adhesion to HPAECs and phenotypic switching to myofibroblast.

Investigating the effects of metals within particulate matter in human alveolar epithelial cells
Conclusion: These novel data show the ability of metals present in PM to modulate a range of signalling pathways to elicit differential cellular responses and activate several potentially pathological mechanisms. Future work will further explore these mechanisms.

COX2 expression is downregulated in a shear stress magnitude-dependent manner in BMPR2-silenced HPAECs: implications for prostacyclin deficiency in PAH
Conclusion: BMPR2 loss may disrupt the flow-driven production of COX2 in HPAECs, which may be an underlying mechanism of endothelial dysfunction in PAH.

Novel strategies for genetic manipulation of human iPSC-derived organoid platforms
Conclusions: Our preliminary data demonstrates modest but successful transfection of patient iPSC-derived organoid cultures ‘in situ’ in 3D matrigel using a commercial transfection reagent. Further development of these methods will facilitate miRNA:Target gene interaction studies to complement ongoing research in iPSC organoid platforms and inform delivery of future therapeutic strategies utilising small RNA.

Role of Connexin 43 increased expression in pulmonary arterial hyperreactivityinduced by the nerve growth factor NGF
In conclusion, NGF pathway is upregulated in IPAH cells. NGF increases Cx43 PA expression through activation of its TrkA receptor. NGF-induced Cx43 increased expression upregulates GAP junction activity, thus promoting PA hyperreactivity.

Thrombin induces stomatin-like protein 2 (STOML2) expression and IL-8/CXCL8 release by ROCK, JNK, and PEA3 pathways in human lung epithelial cells
In this study, we investigate whether Rho kinase (ROCK), JNK, and PEA3 participate in thrombin-induced STOML2 expression and IL-8/CXCL8 release in human lung epithelial cells. We found that thrombin induced increase in STOML2, but not prohibitin, expression in a time-dependent manner. Thrombin also induces STOML2-luciferase activity. Thrombin-induced STOML2 expression and IL-8/CXCL8 release were reduced by PEA3 siRNA. We also found that transfection of A549 cells with PEA3 siRNA reduced PEA3 protein expression. Sirt3-MnSOD signaling pathway attenuates airway epithelial mitochondrial oxidative stress in chronic obstructive pulmonary disease
Conclusions: The data suggest that Sirt3 attenuates CSE-induced mitochondrial oxidative stress in the airway epithelia via deacetylating MnSOD, and may provide a novel therapeutic target for COPD.

Chromatin dynamics via HMGN5 regulates COPD susceptibility
A role for epigenetics in COPD susceptibility is emerging. We previously detected spontaneous emphysema development in HMGN5 deficient mice.
