Up-regulation of C5orf34 promotes lung adenocarcinoma migration and is correlated with worse prognosis.
CONCLUSION: Our results firstly revealed that C5orf34 might play a facilitating role in LAD development and progression by regulating MAPK signaling pathway. Furthermore, our data implied that C5orf34 may be a potential predictor and treatment target for LAD. PMID: 30771479 [PubMed - as supplied by publisher]

Adeno-associated virus 9-mediated RNA interference targeting SOCS3 alleviates diastolic heart failure in rats.
CONCLUSIONS: SOCS3 gene silencing by AAV9-SOCS3 siRNA administration in a DHF rat model significantly reduced myocardial fibrosis and the inflammatory response and improved heart function. Ajuba overexpression regulates mitochondrial potential and glucose uptake through YAP/Bcl-xL/GLUT1 in human gastric cancer.
Abstract Ajuba dysregulation has been reported in several human cancers. However, its expression patterns and biological roles in human gastric cancers have not yet been characterized. In the current study, we found that Ajuba protein was increased in gastric cancer tissues and in cell lines. High Ajuba expression positively correlated with the tumor-node-metastasis (TNM) stage, lymph node metastasis and poor prognosis. The Cancer Genome Atlas (TCGA) and Oncomine microarray data mining also suggested that Ajuba mRNA upregulation in gastric cancer tissues. We used SGC-7901 and NCI-N87 cell lines for Ajuba overexpre...

The role of R-spondin 1 through activating Wnt/ β-catenin in the growth, survival and migration of ovarian cancer cells.
The role of R-spondin 1 through activating Wnt/β-catenin in the growth, survival and migration of ovarian cancer cells. Gene. 2018 Dec 17;: Authors: Liu Q, Zhao Y, Xing H, Li L, Li R, Dai J, Li Q, Fang S Abstract Aberrant activation of the Wnt/β-catenin has been shown to promote progression in various cancers, including ovarian cancer. However, the molecular mechanisms involved in Wnt/β-catenin activation are not well elucidated. In the work, we identify that R-spondin 1 is an upstream regulator in Wnt/β-catenin pathway to promote growth, survival and migration in ovarian cancer cells. We observe ...

Novel role of SF1 in alleviating thyroid-associated ophthalmopathy through the AMPK/mTOR signaling pathway.
CONCLUSION: Our result showed up-regulated SF1 may relieve TAO through suppressing cell proliferation and differentiation, but accelerating cell apoptosis by inhibiting the activation of the AMPK/mTOR signaling pathway. PMID: 30562606 [PubMed - as supplied by publisher]

Preliminary screening and functional analysis of circular RNAs associated with hepatic stellate cell activation.
CONCLUSION: The circRNAs expression profile of liver tissue had changed in fibrosis mouse model, and some of these circRNAs may be associated with HSC activation. For instance, mmu_circ_34116 would inhibit HSC activation. PMID: 30118889 [PubMed - as supplied by publisher]

Activation of p53 by costunolide blocks glutaminolysis and inhibits proliferation in human colorectal cancer cells.
Abstract Colorectal cancer is a leading cause of cancer-related death. Glutaminolysis has been suggested as a therapeutic target for cancer. Costunolide is a natural sesquiterpene lactone showing potent antitumor activity. Our studies were aimed at evaluating how costunolide affected glutaminolysis leading to proliferation inhibition in human colorectal cancer cells. Costunolide suppressed viability and proliferation of HCT116 cells concentration-dependently, but did not apparently affect human intestinal epithelial cells. Costunolide at 20 μM reduced viability and proliferation of HCT116 cells time-dependently...

LncRNA GACAT3 acts as a competing endogenous RNA of HMGA1 and alleviates cucurbitacin B-induced apoptosis of gastric cancer cells.
In this study, we demonstrate that GACAT3 acts as a competing endogenous RNA of high mobility group A1 (HMGA1), a typical oncogene that is overexpressed in most types of cancer, based on a search for common miRNA-binding sites and on experiments involving in vitro cell transfection with synthesized miRNA mimics. Furthermore, knockdown of GACAT3 by its specific siRNA resulted in significantly decreased cell proliferation in gastric cancer cells, similar to the effect of an HMGA1 knockdown. Moreover, GACAT3 overexpression alleviated the apoptosis induced by cucurbitacin B, which is a widely used anticancer drug. Mechanistica...

Loss of histone deacetylase 2 inhibits oxidative stress induced by high glucose via the HO-1/SIRT1 pathway in endothelial progenitor cells.
In conclusion, these findings indicate that an HDAC2 inhibitor may prevent impaired cell proliferation, tube formation, inflammation, and ROS production in high-glucose EPCs. PMID: 30063937 [PubMed - as supplied by publisher]

Long-term evolutionary DNA methylation dynamic of protein-coding genes and its underlying mechanism.
Abstract DNA methylation is an important type of epigenetic modifications for the maintenance of genome functionality and stability. Although there are many studies on DNA methylation patterns, mechanisms, and functions, no study has focused on the evolutionary dynamic of DNA methylation. Here, we present the first genome-wide pattern of evolutionary DNA methylation dynamic in protein-coding genes, by grouping the Arabidopsis thaliana protein-coding genes into several conservation levels representing different evolutionary ages, and by investigating their DNA methylation features for three methylation contexts in ...

LTBP2 knockdown and oxidative stress affect glaucoma features including TGF β pathways, ECM genes expression and apoptosis in trabecular meshwork cells.
LTBP2 knockdown and oxidative stress affect glaucoma features including TGFβ pathways, ECM genes expression and apoptosis in trabecular meshwork cells. Gene. 2018 Jun 13;: Authors: Suri F, Yazdani S, Elahi E Abstract Glaucoma is the leading cause of irreversible blindness worldwide. Although the etiology of glaucoma is incompletely understood, it is known that the extracellular matrix (ECM) of the trabecular meshwork, oxidative stress, TGFβ signaling pathways, and apoptosis are important components of glaucoma pathogenesis. These components appear to be interrelated, but knowledge on their interacti...

MEF2A regulates Calpain 3 expression in L6 myoblasts.
Abstract Calpain 3 (Capn3), a skeletal muscle-specific member of the calpain family, executes some non-proteolytic functions besides its role as a Ca2+-regulated proteolytic enzyme. Previously, we found that changes in Capn3 expression were linearly correlated with the degree of muscular atrophy following reversible sciatic nerve injury and that knockdown of Capn3 gene expression promoted myoblast differentiation. While the regulation of capn3 gene expression is interesting, transcriptional regulation of Capn3 is still unclear. In the present study, we provided experimental data showing that the myogenic enhancer ...

CDH17 alters MMP-2 expression via canonical NF- κB signalling in human gastric cancer.
CDH17 alters MMP-2 expression via canonical NF-κB signalling in human gastric cancer. Gene. 2018 May 18;: Authors: Jiang XJ, Lin J, Cai QH, Zhao JF, Zhang HJ Abstract Gastric cancer (GC), one of the most common cancers of the digestive system, results in high morbidity and mortality, but the molecular mechanisms underlying GC remain largely unknown. Cadherin-17 (CDH17) is a nonclassical member of the cadherin (CDH) superfamily of calcium-dependent proteins. Despite recent advances in the understanding of CDH17 biology, the mechanism of CDH17 in GC proliferation, migration, and invasion has not been e...
