Late Breaking Abstract - DCLK1 mediates thrombin-induced IL-8/CXCL8 expression in lung epithelial cells
Airway neutrophilia has been known correlated with asthma severity. Thrombin plays a crucial role in asthma. Thrombin can induce IL-8/CXCL8 expression, a potent chemoattractant for neutrophils, in lung epithelial cells. Doublecortin-like kinase 1 (DCLK1) is a protein kinase that has been identified as a tumor stem cell marker in colon and pancreatic cancer. Here we investigated the role of DCLK1 in thrombin-induced IL-8/CXCL8 expression in human lung epithelial cells. Thrombin-induced IL-8/CXCL8 release was attenuated by DCLK1 siRNA or LRKK2-IN-1 (DCLK1 inhibitor) in BEAS-2B and A549 cells. Moreover, thrombin-induced IL-8/...

Cbx4 Regulates RNA Binding Protein MEX-3B To Thymic stromal lymphopoietin Expression Sumoylation In Epithelium Of HDM-induced Allergic Inflammation
Conclusion: CBX4/MEX-3B/TSLP axis involved in TSLP-mediated airway inflammation, suggesting that targeting SUMO E3 ligase activity of CBX4 would be a novel target for the treatment of asthma.

Ribosomal protein S3 gene silencing protects against experimental allergic asthma
We reported here for the first time that RPS3 gene silencing ameliorates experimental allergic asthma, probably via interruption of NF-B activity, confirming RPS3 a novel therapeutic target for the treatment of allergic airway inflammation.

NLRP3 inflammasome is activated via phosphoinositide 3-kinase {delta} pathway in Aspergillus fumigatus-induced allergic airway inflammation
In this study, we focused on an association of NLRP3 inflammasome activation with PI3K- signaling in Aspergillus fumigatus (Af)-induced allergic lung inflammation. Using Af-exposed in vivo and in vitro experimental systems, we have investigated the role of PI3K- in the regulation of NLRP3 inflammasome activation in the pathogenesis of fungus-induced allergic lung inflammation. The protein expression of NLRP3, caspase-1, and IL-1bin lungs was significantly increased after the challenge in Af-sensitized and -challenged mice. Protein expression of NLRP3 and caspase-1 was also remarkably elevated in Af-stimulated cultured trach...

LATE-BREAKING ABSTRACT: Inhibition of the ubiquitin-E3-ligase FBXW7 enhances steroid efficacy: New concept for treating steroid insensitivity
Conclusion: Reduction of FBXW7 caused a significant augmentation of functional GR and may have the potential to improve the efficacy of steroids.

CREB/CRTC-mediated regulation of transcription is important for regulatory T cells
Conclusion: Our data point to an activation of CREB/CRTC-mediated transcription in naturally occurring as well as in in vitro induced Tregs. CREB knockdown reduced the formation of iTregs suggesting a possible role of this pathway in immune tolerance.

Expression of Olig2 in terminally differentiated eosinophils among leukocytes
Eosinophils are granulocytes that play a role in homeostasis and pathogenesis of heminth infection and allergic diseases. Olig2, a basic helix loop helix transcription factor, plays a key role in cell fate decision of oligodendrocytes and motor neurons. In the present study, we identified OLIG2 as a gene that among leukocytes is expressed exclusively in eosinophils. When cord blood (CB) CD34+ hematopoietic stem cells differentiated toward eosinophils during a 24 day culture period, Olig2 protein was expressed only in CB eosinophils at day 24, a time when these cells were considered fully differentiated and matured, while O...

LSC 2014 abstract - ROR{gamma} t-specific RNAi decreases allergic airway inflammation and airway hyperresponsiveness in a mouse model of neutrophilic asthma
Conclusion: Our data show that knockdown of ROR t by gene silencing improved airway inflammation in a mouse model of experimental asthma and may have therapeutic potential for the treatment of neutrophilic asthma.

Activation of PI3K-d signaling pathway contributes to the generation of mitochondrial ROS and NLRP3 inflammasome in HDM-inhaled mice
In this study, we have investigated the effects of IC87114, a representative inhibitor of PI3K- isoform, on the HDM-induced airway inflammation and hyperresponsiveness. In addition, we tried to define the role of PI3K- in the HDM-induced NLRP3 inflammasome activation in airways using primary cultured tracheal epithelial cells.HDM-sensitized and -challenged mice showed the typical allergic airway inflammation and hyperresponsivenss, and the increased levels of Th2 cytokines, IL-17, IL-1β, and TNF-α in lungs. Moreover, we found the increases of mitochondrial ROS and the NLRP3 inflammasome activation in HDM-exposed...

Phosphoinositide 3-kinase {delta} signaling modulates endoplasmic reticulum stress in aspergillus fumigatus-induced allergic lung inflammation
In this study, we aimed to elucidate an association of ER stress with PI3K- signaling in fungus-induced allergic lung inflammation.Using Aspergillus fumigatus-exposed in vivo and in vitro experimental systems, we have investigated the role of PI3K- in the regulation of ER stress in the pathogenesis of allergic lung inflammation. The Aspergillus fumigatus-sensitized and -challenged mice showed that the expression of ER stress markers and the protein levels of unfolded protein response (UPR)-related markers in lung tissues were significantly increased after Aspergillus fumigatus challenge. Protein expression of ER stress mark...
