Cyclo-(Phe-Tyr) as a novel cyclic dipeptide compound alleviated ischemic stroke reperfusion brain injury via JUNB/JNK/NF- κB and SOX5/PI3K/AKT pathways
Pharmacol Res. 2022 Apr 25:106230. doi: 10.1016/j.phrs.2022.106230. Online ahead of print.ABSTRACTIschemic stroke reperfusion (IR) can cause adverse reactions including apoptosis, oxidative stress, and inflammation, but the existing therapeutic strategies have been limited. Moreover, the regulation of microglia plays an important role in brain injury after reperfusion. Hence, it is imperative to find new and effective drugs for modulating microglia to treat IR brain injury. Cyclic peptide compound cyclo-(Phe-Tyr) (Sparganin C, SC) is a compound isolated from Sparganii Rhizoma. However, the protective effects of SC on the c...

Inhibition of the immunoproteasome LMP2 ameliorates ischemia/hypoxia-induced blood-brain barrier injury through the Wnt/ β-catenin signalling pathway
CONCLUSION: This study suggests that inhibition of the immunoproteasome LMP2 ameliorates ischemia/hypoxia-induced BBB injury, and that the molecular mechanism involves the immunoproteasome-regulated activation of the Wnt/β-catenin signalling pathway under ischemic conditions.PMID:34857032 | DOI:10.1186/s40779-021-00356-x

MiR-499a prevents astrocytes mediated inflammation in ischemic stroke by targeting PTEN
CONCLUSIONS: Taken together, our findings indicate miR-499a as an important factor to prevent inflammatory response and suggest miR-499a as a new molecule for the treatment of IS. The present study also demonstrated the relationship between miR-499a and PTEN, with PTEN as a downstream signaling mediator of miR-499a in the inflammatory response of astrocytes induced by IS.PMID:34632168 | PMC:PMC8488463 | DOI:10.1016/j.ncrna.2021.09.002

Expression of Long Non-coding RNA RGD1566344 in the Brain Cortex of Male Mice After Focal Cerebral Ischemia-Reperfusion and the Neuroprotective Effect of a Non-coding RNA RGD1566344 Inhibitor.
In this study, we found that RGD1566344 levels were elevated in the ischemic infarct and penumbra regions 12 h after middle cerebral artery occlusion/reperfusion (MCAO/R) in male mice and in PC12 cells with oxygen glucose deprivation/reperfusion (OGD/R). The inhibition of RGD1566344 by small interference RNA (siRNA) significantly alleviated apoptosis in OGD/R PC12 cells. In cell transfection, quantitative real-time PCR, and Western blot experiments, we demonstrated the possible interaction of non-POU domain-containing octamer-binding protein (NONO) with RGD1566344. The NONO level in OGD/R PC12 cells was obviously increase...

Upregulation of NOX2 and NOX4 Mediated by TGF- β Signaling Pathway Exacerbates Cerebral Ischemia/Reperfusion Oxidative Stress Injury
Conclusions: Our studies demonstrated that TGF- β signaling activation is involved in the regulation of NOX2/NOX4 expression and exacerbates cerebral I/R injury.Cell Physiol Biochem 2018;46:2103 –2113

IL-8 enhances angiogenesis of human bone marrow mesenchymal stem cells by increasing vascular endothelial growth factor.
In conclusion, IL-8 stimulates VEGF production in hBM-MSCs in part via the PI3K/Akt and MAPK/ERK signal transduction pathways and that administration of IL-8-treated hBM-MSCs increases angiogenesis after stroke. This approach may be used to optimize MSC-based therapies for numerous diseases including stroke, myocardial ischemia, and spinal cord injury. PMID: 24797366 [PubMed - as supplied by publisher]
