Knocking down SOX2 overcomes the resistance of prostate cancer to castration via notch signaling
CONCLUSION: We demonstrated that SOX2/Notch signaling was responsible for Enzalutamide resistance in CRPC. Targeting SOX2/Notch signaling might represent a new choice for the treatment and therapy of CRPC.PMID:37716921 | DOI:10.1007/s11033-023-08757-y

Knockdown of LIMD2 inhibits the progression of ovarian carcinoma through ERK1/2 pathway
CONCLUSION: Knockdown of LIMD2 inhibits cell proliferation, migration, invasion and autophagy, and promotes the apoptosis through the ERK1/2 signaling pathway, suggesting that LIMD2-siRNA may be an effective molecule to prevent OC progression.PMID:37716918 | DOI:10.1007/s11033-023-08733-6

Nobiletin regulates invasion, migration and metastasis in hypopharyngeal squamous cell carcinoma
This study examined the effects of NOB on the proliferation, migration, and invasiveness of the FaDu cell line derived from hypopharyngeal squamous cell carcinoma. We determined protein kinase phosphorylation by western blot analysis, migration by Transwell assay, and metastatic potential by enzyme-linked immunosorbent assay of vascular endothelial growth factor. NOB inhibited cell proliferation by 40% at concentrations of 15 µM and 40 µM, and reduced migration and induced apoptosis at 50 µM by downregulating extracellular signal-regulated kinase, protein kinase B, and phosphoinositide 3-kinase. Our results suggest that...

The linkage between microRNA and cancer and its delivery as cancer therapy: A mini-review
Cell Mol Biol (Noisy-le-grand). 2023 Jul 31;69(7):7-18. doi: 10.14715/cmb/2023.69.7.2.ABSTRACTThe central dogma of molecular biology was no longer "central" after ground-breaking discoveries conveyed gene expression involves more complex physiological functions in cancer pathogenesis over the last decade. MicroRNAs (miRNAs) are short non-coding RNA that regulate gene expression, affecting key molecular pathways involved in sustaining the proliferative signalling for tumour development, evasion of cellular death, invasion, angiogenesis, as well as metastasis in a plethora of cancer types. MiRNA expression is dysregulated in...

Molecular Subtypes Based on PANoptosis Genes and Characteristics of Immune Infiltration in Cutaneous Melanoma
Cell Mol Biol (Noisy-le-grand). 2023 Aug 31;69(8):1-8. doi: 10.14715/cmb/2023.69.8.1.ABSTRACTCutaneous melanoma (CM) remains the most life-threatening form of skin cancer. Further risk stratification and search for new prognostic targets for CM are of positive clinical significance. PANoptosis is defined as an inflammatory programmed cell death mediated by the PANoptosome complex and cannot be characterized by pyroptosis, apoptosis or necroptosis alone. Although PANoptosis is closely associated with many diseases including cancer, it has not been reported in CM. Combined with GTEx and TCGA database, we extracted 14 PANopto...

CRISPR/Cas-9 vector system: targets UL-39 and inhibits Simplexvirus humanalpha1 (HSV-1) replication in vitro
Cell Mol Biol (Noisy-le-grand). 2023 Jul 31;69(7):19-23. doi: 10.14715/cmb/2023.69.7.3.ABSTRACTSimplexvirus humanalpha1 (HSV-1) affects approximately 67% of the world's population. Here, we sought to use the CRISPR / Cas9 system with the UL39 target, essential for virus replication. The sgRNA sequence was inserted into the plasmid (PX459-UL39). Vero cells were transfected with PX459-UL39, and inhibition of viral replication was assessed 24 and 48 hours later using plaque assays and fluorescence and qPCR. Fluorescence analyses revealed the presence of anti-HSV-1 CRISPR/Cas9 within Vero cells, and qPCR showed that the vira...

Vaginal Microbiota Profile in first-trimester miscarriages cases
In this study, 435 vaginal swabs were collected from aborted women in Maternity Teaching Hospitals in Erbil and Shahid Dr. Khalid Hospital in Koya. We characterized the vaginal microbiota diversity and composition in first-trimester abortion and investigated the association between bacterial vaginosis and abortion before 12 weeks. Cultural, morphological, and biochemical characteristics for each bacterial genus were discovered, and VITEK-2 system was used to identify isolated bacteria. Samples from each type of bacteria were selected for sequencing utilizing 16 rRNA genes examining V4-V8 region for bacterial profiles. Bact...

Recombinant Antibodies to Face SARS-CoV-2 Syndemic
Cell Mol Biol (Noisy-le-grand). 2023 Jul 31;69(7):24-27. doi: 10.14715/cmb/2023.69.7.4.ABSTRACTIn recent years, increasing interest has been paid to using antibody-based therapies for clinical applications. However, it is unclear whether recombinant antibodies can be combined with other scientific approaches to generate innovative solutions for mitigating severe acute respiratory syndrome coronavirus 2. In this context, the increase in this virus transmission, the number of infected people, and the interaction between social and biological processes have led to a syndemic, exacerbating the public health problem. Here, we a...

Resveratrol alleviates MSU-induced gouty arthritis in rats through inhibition of HIF-1 α- and NLRP3-derived IL-1β secretion in macrophages
This study demonstrated that resveratrol significantly improved the symptoms of acute gouty arthritis and its potential mechanism may be IL-1β reduction via HIF-1α modulation and inhibition of NLRP3 inflammasome activation. Our study might offer a novel sight for the treatment of gouty arthritis.PMID:37715438 | DOI:10.14715/cmb/2023.69.7.5

The role of galectin-3 in patients with permanent and paroxysmal Atrial Fibrillation and echocardiographic parameters of left atrial fibrosis
CONCLUSIONS: Echocardiographic findings showed association with the groups, and with serum levels of Gal-3 in patients with permanent AF. The distribution of allelic and genotypic frequencies, and of the haplotypes of polymorphism LGALS3 rs4652 and rs4644 did not present statistical variation, which suggests that those SNPs are not associated with the AF clinical forms (permanent and paroxysmal).PMID:37716919 | DOI:10.1007/s11033-023-08774-x

Constructing phylogenetic networks via cherry picking and machine learning
CONCLUSIONS: Unlike the existing exact methods, our heuristics are applicable to datasets of practical size, and the experimental study we conducted on both simulated and real data shows that these solutions are qualitatively good, always within some small constant factor from the optimum. Moreover, our machine-learned heuristics are one of the first applications of machine learning to phylogenetics and show its promise.PMID:37717003 | DOI:10.1186/s13015-023-00233-3
