Elucidating the morphogenic and signaling roles of defined growth factors controlling human endothelial cell lumen formation versus sprouting behavior
Am J Pathol. 2023 Sep 7:S0002-9440(23)00317-6. doi: 10.1016/j.ajpath.2023.08.009. Online ahead of print.ABSTRACTWe have previously shown that five growth factors (Factors) [i.e. insulin, fibroblast growth factor-2 (FGF-2), stem cell factor (SCF), interleukin-3 (IL-3), and stromal-derived factor-1 alpha (SDF-1α)] in combination are necessary for human ECs to undergo tube morphogenesis, a process requiring both lumen formation and sprouting behavior. Here, we investigate why they are required by subdividing the Factors into 4 separate groups: Insulin only, Insulin + FGF-2, no FGF-2 (all Factors but without FGF-2), and all F...

Defective uterine spiral artery remodelling and placental senescence in a pregnant rat model of polycystic ovary syndrome
This study verified previous work in which exposure to 5-dihydrotestosterone (DHT) and insulin (INS) in rats during pregnancy resulted in hyperandrogenism, insulin intolerance, and higher foetal mortality. It also found that exposure to DHT and INS dysregulated the expression of angiogenesis-related genes in the uterus and placenta. It also decreased expression of endothelial nitric oxide synthase and matrix metallopeptidases 2 and 9, increased fibrotic collagen deposits in the uterus, and reduced expression of marker genes for SpA-associated trophoblast giant cells. These changes were related to a greater proportion of un...

Dolutegravir use over 48 weeks is not associated with worsening insulin resistance and pancreatic beta cell function in a cohort of HIV-infected Ugandan adults
CONCLUSION: We demonstrated insignificant changes in both insulin resistance and pancreatic beta cell function in clinically stable young adult Ugandan PWH on dolutegravir for 48 weeks. We add to the body of evidence demonstrating glucose metabolic safety of dolutegravir in ART naïve patients. Ugandan guidelines should reconsider restricting DTG initiation in ART naive adults at high risk for diabetes.PMID:37689695 | PMC:PMC10492310 | DOI:10.1186/s12981-023-00564-6

Probing the molecular interactions between cholinium-based ionic liquids and insulin aspart: A combined computational and experimental study
Int J Biol Macromol. 2023 Sep 7:126665. doi: 10.1016/j.ijbiomac.2023.126665. Online ahead of print.ABSTRACTDespite extensive studies revealing the potential of cholinium-based ionic liquids (ILs) in protein stabilization, the nature of interaction between ILs' constituents and protein residues is not well understood. In this work, we used a combined computational and experimental approach to investigate the structural stability of a peptide hormone, insulin aspart (IA), in ILs containing a choline cation [Ch]+ and either dihydrogen phosphate ([Dhp]-) or acetate ([Ace]-) as anions. Although IA remained stable in both 1 M [C...
