Modulation of paraoxonase-2 in human dermal fibroblasts by UVA-induced oxidative stress: A new potential marker of skin photodamage
Chem Biol Interact. 2023 Sep 15:110702. doi: 10.1016/j.cbi.2023.110702. Online ahead of print.ABSTRACTParaoxonase-2 (PON2) is an intracellular protein widely expressed in many cell types and tissues, whose levels are known to be modulated by oxidative stress and apoptosis induced by genetic and environmental factors. However, an environmental factor that has not been previously explored is the effect of ultraviolet A radiation (UVA), the oxidizing component of sunlight, on the modulation of PON2 in human dermal fibroblasts (HuDe). Excessive UVA radiation is known to cause direct and indirect skin damage by influencing intr...

Nobiletin regulates invasion, migration and metastasis in hypopharyngeal squamous cell carcinoma
This study examined the effects of NOB on the proliferation, migration, and invasiveness of the FaDu cell line derived from hypopharyngeal squamous cell carcinoma. We determined protein kinase phosphorylation by western blot analysis, migration by Transwell assay, and metastatic potential by enzyme-linked immunosorbent assay of vascular endothelial growth factor. NOB inhibited cell proliferation by 40% at concentrations of 15 µM and 40 µM, and reduced migration and induced apoptosis at 50 µM by downregulating extracellular signal-regulated kinase, protein kinase B, and phosphoinositide 3-kinase. Our results suggest that...

Molecular Subtypes Based on PANoptosis Genes and Characteristics of Immune Infiltration in Cutaneous Melanoma
Cell Mol Biol (Noisy-le-grand). 2023 Aug 31;69(8):1-8. doi: 10.14715/cmb/2023.69.8.1.ABSTRACTCutaneous melanoma (CM) remains the most life-threatening form of skin cancer. Further risk stratification and search for new prognostic targets for CM are of positive clinical significance. PANoptosis is defined as an inflammatory programmed cell death mediated by the PANoptosome complex and cannot be characterized by pyroptosis, apoptosis or necroptosis alone. Although PANoptosis is closely associated with many diseases including cancer, it has not been reported in CM. Combined with GTEx and TCGA database, we extracted 14 PANopto...

Cell Mol Biol (Noisy-le-grand). 2023 Jul 31;69(7):19-23. doi: 10.14715/cmb/2023.69.7.3.ABSTRACTSimplexvirus humanalpha1 (HSV-1) affects approximately 67% of the world's population. Here, we sought to use the CRISPR / Cas9 system with the UL39 target, essential for virus replication. The sgRNA sequence was inserted into the plasmid (PX459-UL39). Vero cells were transfected with PX459-UL39, and inhibition of viral replication was assessed 24 and 48 hours later using plaque assays and fluorescence and qPCR. Fluorescence analyses revealed the presence of anti-HSV-1 CRISPR/Cas9 within Vero cells, and qPCR showed that the vira...
