A rapid and simple protocol for concentration of SARS-CoV-2 from sewage
J Virol Methods. 2021 Aug 26;297:114272. doi: 10.1016/j.jviromet.2021.114272. Online ahead of print.ABSTRACTThe aim of this study was to set up a simple protocol to concentrate SARS-CoV-2 from sewage, which can be implemented in laboratories with minimal equipment resources. The method avoids the need for extensive purification steps and reduces the concentration of potential inhibitors of RT-qPCR contained in sewage. The concentration method consists of a single step, in which a small volume (40 mL) of sewage sample is incubated with polyaluminum chloride (PAC)(0.00045 N Al3+ final concentration). Virus particles adsorbed...

Systemic administration of choline acetyltransferase decreases blood pressure in murine hypertension
Mol Med. 2021 Oct 21;27(1):133. doi: 10.1186/s10020-021-00380-6.ABSTRACTAcetylcholine (ACh) decreases blood pressure by stimulating endothelium nitric oxide-dependent vasodilation in resistance arterioles. Normal plasma contains choline acetyltransferase (ChAT) and its biosynthetic product ACh at appreciable concentrations to potentially act upon the endothelium to affect blood pressure. Recently we discovered a T-cell subset expressing ChAT (TChAT), whereby genetic ablation of ChAT in these cells produces hypertension, indicating that production of ACh by TChAT regulates blood pressure. Accordingly, we reasoned that incre...

Allergic reactions to COVID-19 vaccines and addressing vaccine hesitancy: Northwell Health experience
CONCLUSION: Intradermal PEG testing has limited utility in evaluating anaphylaxis to mRNA vaccines. Most severe postvaccination allergic symptoms are not caused by hypersensitivity to PEG. The majority of people with reaction to the initial mRNA vaccine can be safely revaccinated. Patients with anaphylaxis to COVID-19 vaccines benefit from physician-observed vaccination.PMID:34699968 | PMC:PMC8542398 | DOI:10.1016/j.anai.2021.10.019

Utility and Futility of Skin Testing to Address Concerns Surrounding mRNA COVID-19 Vaccine Reactions
CONCLUSION: The low positivity rate of COVID-19 vaccine excipient skin testing and high rate of subsequent COVID-19 vaccine tolerance suggests a low utility of this method in evaluation of COVID-19 vaccine hypersensitivity reactions. Focus should shift to the use of existing vaccine allergy practice parameters, with consideration of graded dosing if necessary. Based on these results, strict avoidance of subsequent COVID-19 vaccination should be discouraged.PMID:34798275 | PMC:PMC8594060 | DOI:10.1016/j.anai.2021.11.006

Production of Long-Acting CNGRC-CPG2 Fusion Proteins: New Derivatives to Overcome Drug Immunogenicity of Ligand-Directed Enzyme Prodrug Therapy for Targeted Cancer Treatment
Conclusion: We have successfully produced PEGylated-CNGRC-CPG2, which is bioactive and with lower immunogenicity in ligand-directed enzyme prodrug therapy for cancer treatment.PMID:34802309 | PMC:PMC8606725 | DOI:10.1177/15330338211057371

Factors influencing recovery of SARS-CoV-2 RNA in raw sewage and wastewater sludge using polyethylene glycol-based concentration method
In conclusion, our modified PEG-based concentration method can cut down the analysis time at least by half, which in turn helps to implement early detection system for SARS-CoV-2 in wastewater.PMID:35027874 | PMC:PMC8730514 | DOI:10.7171/jbt.21-3203-012

Biocompatible chitosan/polyethylene glycol/multi-walled carbon nanotube composite scaffolds for neural tissue engineering
J Zhejiang Univ Sci B. 2022 Jan 15;23(1):58-73. doi: 10.1631/jzus.B2100155.ABSTRACTCarbon nanotube (CNT) composite materials are very attractive for use in neural tissue engineering and biosensor coatings. CNT scaffolds are excellent mimics of extracellular matrix due to their hydrophilicity, viscosity, and biocompatibility. CNTs can also impart conductivity to other insulating materials, improve mechanical stability, guide neuronal cell behavior, and trigger axon regeneration. The performance of chitosan (CS)/polyethylene glycol (PEG) composite scaffolds could be optimized by introducing multi-walled CNTs (MWCNTs). CS/PEG...
