A polydopamine nanomedicine used in photothermal therapy for liver cancer knocks down the anti-cancer target NEDD8-E3 ligase ROC1 (RBX1)
Knocking down the oncogene ROC1 with siRNA inhibits the proliferation of cancer cells by suppressing the Neddylation pathway. However, methods for delivering siRNA in vivo to induce this high anticancer activi...

DNA Origami Frameworks Enabled Self ‐Protective siRNA Delivery for Dual Enhancement of Chemo‐Photothermal Combination Therapy
Octahedral DNA origami frameworks (OctDOFs) are constructed for sterically enveloping and efficiently delivering siRNAs for chemo-photothermal combination cancer therapy. The confined space created by this nanoscaffold is able to shield the inward anchored siRNAs from degradative RNases during blood circulation, thus achieved dual sensitization of cancer cells to chemodrugs and hyperthermia. AbstractAlthough chemotherapy and photothermal therapy are widely used to combat cancer, their efficacy is often limited by multidrug resistance. Small interfering RNAs (siRNAs) have ability to suppress the expression of target genes, ...

Modular Lipid Nanoparticle Platform Technology for siRNA and Lipophilic Prodrug Delivery
Lipid nanoparticles (LNPs) are the most advanced nanocarriers for nucleic acid therapeutics such as small interfering RNA (siRNA). Here, it is showcased how LNP technology's modularity and the ability to stably incorporate lipophilic small molecule prodrugs provide ample opportunities to develop combination therapies by co-encapsulating multiple therapeutic agents in a single formulation. AbstractSuccessfully employing small interfering RNA (siRNA) therapeutics requires the use of nanotechnology for efficient intracellular delivery. Lipid nanoparticles (LNPs) have enabled the approval of various nucleic acid therapeutics. ...

Delivery of siHIF ‐1α to Reconstruct Tumor Normoxic Microenvironment for Effective Chemotherapeutic and Photodynamic Anticancer Treatments
An integrated amphiphilic delivery system (RSCD) based on Angiotensin II receptor blocker candesartan for siRNA against the hypoxia-inducible factor-1 alpha (HIF-1 α) is fabricated, aiming at both vascular and cellular “relaxation” to reconstruct a tumor normoxic microenvironment, which significantly improves the chemotherapeutic and photodynamic anticancer treatment. AbstractThe tumor hypoxic microenvironment not only induces genetic and epigenetic changes in tumor cells, immature vessels formation for oxygen demand, but also compromises the efficiency of therapeutic interventions. On the other hand, conventional the...

Therapeutic Gene Silencing Using Targeted Lipid Nanoparticles in Metastatic Ovarian Cancer
Lipid nanoparticles encapsulating small interfering ribonucleic acids are developed that block two essential pathways for cancer cell survival. Targeting of metastasized ovarian cancer cells is further enhanced by coating nanoparticles with hyaluronan. Robust gene silencing in key tissues involved in ovarian cancer pathophysiology in an advanced ovarian cancer mouse model is demonstrated and improved median and overall survival observed. AbstractOvarian cancer is an aggressive tumor owing to its ability to metastasize from stage II onward. Herein, lipid nanoparticles (LNPs) that encapsulate combination of small interfering...
