Hesperidin ameliorates impairment in hippocampal neural stem cells related to apoptosis induced by methotrexate in adult rats
This study investigated whether Hsd could attenuate impairments of hippocampal neural stem cells related to apoptosis induced by MTX. Spraque-Dawley rats (n = 24) were divided into 4 groups: (1) Vehicle group received propylene glycol (21 days) and 0.9% normal saline (day 8 and 15), (2) Hsd group received 100 mg/kg (21 days), (3) MTX group received 75 mg/kg (days 8 and 15) and (4) MTX+Hsd group received MTX, 75 mg/kg (day 8 and 15) and Hsd 100 mg/kg (21 days). Our results showed that MTX decreased hippocampal neural stem cells including SRY (sex determining region Y)-box 2 (SOX2) and nestin. MTX diminished vascular related...

Spermatogonial Dedifferentiation into Germline Stem Cells in Drosophila Testes
Methods Mol Biol. 2023;2677:139-149. doi: 10.1007/978-1-0716-3259-8_8.ABSTRACTStem cell pools are dynamic and capable of reacting to insults like injury and starvation. Recent work has highlighted the key role of dedifferentiation as a conserved mechanism for replenishing stem cell pools after their loss, thereby maintaining tissue homeostasis. The testis of the fruit fly Drosophila melanogaster offers a simple but powerful system to study dedifferentiation, the process by which differentiating spermatogonia can revert their fate to become fully functional germline stem cells (GSCs). Dedifferentiated GSCs show interesting ...

Enhanced osteogenesis on proantocyanidin-loaded date palm endocarp cellulosic matrices: A novel sustainable approach for guided bone regeneration
Int J Biol Macromol. 2023 May 13:124857. doi: 10.1016/j.ijbiomac.2023.124857. Online ahead of print.ABSTRACTDeveloping inexpensive, biocompatible natural scaffolds that can support the differentiation and proliferation of stem cells has been recently emphasized by the research community to faster obtain the FDA approvals for regenerative medicine. In this regard, plant-derived cellulose materials are a novel class of sustainable scaffolding materials with high potentials for bone tissue engineering (BTE). However, low bioactivity of the plant-derived cellulose scaffolds restricts cell proliferation and cell differentiation...

Notch signaling sculpts the stem cell niche
Front Cell Dev Biol. 2022 Dec 20;10:1027222. doi: 10.3389/fcell.2022.1027222. eCollection 2022.ABSTRACTAdult stem cells depend on their niches for regulatory signaling that controls their maintenance, division, and their progeny differentiation. While communication between various types of stem cells and their niches is becoming clearer, the process of stem cell niche establishment is still not very well understood. Model genetic organisms provide simplified systems to address various complex questions, for example, how is a stem cell niche formed? What signaling cascades induce the stem cell niche formation? Are the mecha...

Eleven science stories likely to make big news in 2023
As the COVID-19 pandemic enters its fourth year as a global health emergency, researchers will continue pushing to help make the disease manageable and ordinary. They will track hundreds of subvariants of Omicron, the highly transmissible but seemingly less lethal strain of SARSCoV-2 that dominated in 2022. Virologists will watch the virus’ evolution this year to see whether it has finally slowed or a more dangerous variant pops up, evading much of the immunity that humanity has built up to previous ones. Vaccine researchers hope to develop new shots that provide broad protection against a variety of coronaviruses.  Ano...

Toosendanin inhibits colorectal cancer cell growth through the Hedgehog pathway by targeting Shh
Drug Dev Res. 2022 Jun 2. doi: 10.1002/ddr.21951. Online ahead of print.ABSTRACTColorectal cancer (CRC) is one of the most common gastrointestinal cancers worldwide. This complex and often fatal disease has a high mortality rate. The Hedgehog (Hh) signaling pathway is crucial in CRC. Many studies have indicated that Shh is overexpressed in cancer stem cells (CSCs), and shh overexpression is positively correlated with CRC tumorigenesis. New drugs that kill CRC cells through the Hh pathway are needed. Toosendanin (TSN), a natural triterpenoid saponin extracted from the bark or fruit of Melia toosendan Sieb. et Zucc, can inhi...
