Identification of healthspan-promoting genes in Caenorhabditis elegans based on a human GWAS study
Biogerontology. 2022 Jun 24. doi: 10.1007/s10522-022-09969-8. Online ahead of print.ABSTRACTTo find drivers of healthy ageing, a genome-wide association study (GWAS) was performed in healthy and unhealthy older individuals. Healthy individuals were defined as free from cardiovascular disease, stroke, heart failure, major adverse cardiovascular event, diabetes, dementia, cancer, chronic obstructive pulmonary disease (COPD), asthma, rheumatism, Crohn's disease, malabsorption or kidney disease. Six single nucleotide polymorphisms (SNPs) with unknown function associated with ten human genes were identified as candidate healths...

A suggested shared aetiology of dementia - a colocalization study
Neurobiol Aging. 2022 May 13;117:71-82. doi: 10.1016/j.neurobiolaging.2022.05.005. Online ahead of print.ABSTRACTIdentification of shared causal genes between dementia and its related clinical outcomes can help understand shared aetiology and multimorbidity surrounding dementia. We performed the HyPrColoc colocalization analysis to detect possible shared causal genes between dementia or Alzheimer's disease (AD) and 5 selected traits: stroke, diabetes, atherosclerosis, cholesterol level, and alcohol consumption within 601 dementia or AD associated genetic regions using summary results of the UK Biobank genome-wide associati...

Imaging subtle leaks in the blood –brain barrier in the aging human brain: potential pitfalls, challenges, and possible solutions
AbstractRecent studies using dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI) with gadolinium-based contrast agents (GBCA) have demonstrated subtle blood –brain barrier (BBB) leaks in the human brain during normal aging, in individuals with age-related cognitive dysfunction, genetic risk for Alzheimer’s disease (AD), mild cognitive impairment, early AD, cerebral small vessel disease (SVD), and other neurodegenerative disorders. In these neurologi cal conditions, the BBB leaks, quantified by the unidirectional BBB GBCA tracer’s constantKtrans maps, are typically orders of magnitude lower than in brain tu...

Association between Baseline Cognitive Function and Longitudinal Functional Outcome Change after Ischemic Stroke
Conclusion: After ischemic stroke, higher baseline global cognitive function was a predictive factor for better functional recovery regardless of depression symptoms in the minor stroke group.Dement Geriatr Cogn Disord

Impact of Pre-Stroke Dementia or Mild Cognitive Impairment on Stroke Outcome: A Systematic Review and Meta-Analysis
Conclusions: Our meta-analysis shows an increased risk of mortality in stroke patients with a history of PSD and PSMCI. Proper clinical management and increased attention are therefore required for the prevention and management of stroke in patients with cognitive deficits.Dement Geriatr Cogn Disord

Longitudinal white matter hyperintensity changes and cognitive decline in patients with minor stroke
CONCLUSIONS: The longitudinal change of WMH is dynamic. The regressive WMH volume was associated with the use of antihypertensive medications on a regular basis. The presence of lacunes at the initial visit of the study was a stronger predictor of WMH progression. The progression of WMH volume could be useful in predicting cognitive decline in patients with minor stroke.PMID:35084664 | DOI:10.1007/s40520-021-02024-5

Cardiometabolic and Vascular Disease Factors and Mild Cognitive Impairment and Dementia
Conclusion: Older adults with higher burden of CMVD, driven especially by diabetes, are likely to increase the risk of prevalent and incident MCI and dementia.Gerontology

The health status: the ignored risk factor in dementia incidence. NEDICES cohort
CONCLUSIONS: Our data indicate that it is worthwhile to include the SPH status as a RF in the studies of dementia and AD incidence and to explore the effect of its improvement in the evolution of this incidence.PMID:35025095 | DOI:10.1007/s40520-021-02045-0

Is There an Association between Norton Scale and Cognitive Impairment in Hip Fractured Elderly Patients?
Conclusion: Our findings suggest that there is an association between Norton Scale Scores and cognitive impairment. Norton score parameters, under certain circumstances, such as speech and other communication difficulties, can be used as a proxy measure for MMSE to indicate cognitive impairment. These findings can be even more helpful in the present time of “COVID-19,” when we have to evaluate older patients with facial masks and other defensive suits.Dement Geriatr Cogn Disord

Leukocyte-derived ratios are associated with late-life any type dementia: a cross-sectional analysis of the Mugello study
AbstractImmunosenescence, vascular aging, and brain aging, all characterized by elevated levels of inflammatory markers, are thought to share a common pathogenetic pathway: inflamm-aging. Retrospective cross-sectional analysis was conducted using data from the Mugello study (Tuscany, Italy), a representative Italian cohort of free-living nonagenarians. to assess the association between specific peripheral inflammation markers derived from white blood cell counts, and the diagnosis of dementia. All the variables of interest were reported for 411 subjects (110 males and 301 females) out of 475 enrolled in the study. Anamnest...
