A Novel Paradigm Defines Functional Molecule Clusters for an Anti-Alzheimer's Disease Recipe from Traditional Chinese Medicine
In this study, a similarity analysis based on molecular fingerprints was adopted to explore the representative molecules of the Tiaoxin recipe, a Chinese patent formula approved by the National Medical Products Administration (NMPA) for the treatment of mild-to-moderate Alzheimer's disease (AD), and 38 out of 1047 chemicals were finally screened out. Next, we tried to define a new concept of a "functional molecule cluster" for chemicals with similar pharmacological effects to elucidate how the chemical mixture from TCMs produce their therapeutic effects. Four anti-AD functional molecule clusters from the Tiaoxin recipe wer...

Don ’ t Get Dragged Down By “ Zombie Cells ”
Did you watch any of the zombie shows that were all over the airways a few years back? I confess my sons and I really got into the program The Walking Dead. Watching the cast of characters try to survive in a world overrun by dead zombies was a fun way to spend some time together and wrap up a Sunday night. Typically, when I think of the word “zombie,” I’m thinking of science fact, not fiction… Zombie cells are known scientifically as senescent cells. These are dysfunctional cells that have stopped dividing and are nearing the end of their natural lives. Zombie cells are too sick and damaged to carry out the...

Constituent isoflavones of Puerariae radix as a potential neuroprotector in cognitive impairment: Evidence from preclinical studies
Ageing Res Rev. 2023 Aug 23;90:102040. doi: 10.1016/j.arr.2023.102040. Online ahead of print.ABSTRACTWith the increasing aging population worldwide, the incidence of senile cognitive impairment (CI) is increasing, posing a serious threat to the health of elderly persons. Despite developing new drugs aimed at improving CI, progress in this regard has been insufficient. Natural preparations derived from plants have become an unparalleled resource for developing new drugs. Puerariae radix (PR) has a long history as Chinese herbal medicine. PR is rich in various chemical components such as isoflavones, triterpenes, and saponin...

Isoniazid derivatization strategy of carboxyl-containing metabolites for LC-MS/MS-based targeted metabolomics
Anal Bioanal Chem. 2023 Aug 25. doi: 10.1007/s00216-023-04910-5. Online ahead of print.ABSTRACTMetabolomics is a biochemical analysis tool for identifying metabolic phenotypes and used to reveal the pathogenic mechanisms of disease and to inform drug-targeted therapies. Carboxyl-containing metabolites (CCMs) account for an important proportion of the metabolome, but because of the diversity of physical and chemical properties of CCMs in biological samples, traditional liquid chromatography-mass spectrometry (LC-MS) targeted metabolome analysis methods cannot achieve simultaneous quantification of multiple types of CCMs. Th...

Levistilide A ameliorates neuroinflammation via inhibiting JAK2/STAT3 signaling for neuroprotection and cognitive improvement in scopolamine-induced Alzheimer's disease mouse model
Int Immunopharmacol. 2023 Aug 22;124(Pt A):110783. doi: 10.1016/j.intimp.2023.110783. Online ahead of print.ABSTRACTAlzheimer's disease (AD) is a progressive neurodegenerative disease associated with cognitive impairment and dementia, which has become a major public health problem. There are no effective therapeutic agents used to treat AD in clinic for the extremely complex pathogenesis. Here we identify Levistilide A (LA), one of the major active natural terpene lactone constituents from Chinese herbal medicine Angelicae sinensis and Chuanxiong Rhizoma, as a potent neuroinflammation inhibitor for neuroprotection and cogn...

Research on the Chemical Constituents against Alzheimer's Disease of the Fruits of Physalis alkekengi L. var. franchetii (Mast.) Makino
This study reveals that PAF can reduce neuroinflammatory damage by inhibiting p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway, which is the theoretical basis for clinical development and utilization of PAF in AD therapy.PMID:37505462 | DOI:10.1002/cbdv.202301075

The Role of monosodium glutamate (MSG) in Epilepsy and other Neurodegenerative Diseases: Phytochemical-based Therapeutic Approaches and Mechanisms
This study is focused on neuroprotective herbal medications that can pass the blood-brain barrier and cure MSG-induced NDDs and the factors that influence MSG-induced glutaminergic, astrocyte, and GABAergic neuron abnormalities causing neurodegeneration.PMID:37496245 | DOI:10.2174/1389201024666230726161314
