< em > hGFAP < /em > -mediated < em > GLI2 < /em > overexpression leads to early death and severe cerebellar malformations with rare tumor formation
iScience. 2023 Jul 28;26(9):107501. doi: 10.1016/j.isci.2023.107501. eCollection 2023 Sep 15.ABSTRACTThe zinc-finger transcription factor GLI2 is frequently amplified in childhood medulloblastoma of the Sonic-hedgehog type (SHH-MB), with or without amplification of NMYC or deletion of TP53. Despite the aggressive tumor behavior, tumorigenesis is not well understood, and adequate mouse models are lacking. Therefore, we generated mice with a GLI2 overexpression under control of the hGFAP-promoter. These mice died within 150 days. The majority only survived until postnatal day 40. They displayed severe cerebellar hypoplasia, ...

Mutation detection of urinary cell-free DNA via catch-and-release isolation on nanowires for liquid biopsy
Biosens Bioelectron. 2023 Apr 23;234:115318. doi: 10.1016/j.bios.2023.115318. Online ahead of print.ABSTRACTCell-free DNA (cfDNA) and extracellular vesicles (EVs) are molecular biomarkers in liquid biopsies that can be applied for cancer detection, which are known to carry information on the necessary conditions for oncogenesis and cancer cell-specific activities after oncogenesis, respectively. Analyses for both cfDNA and EVs from the same body fluid can provide insights into screening and identifying the molecular subtypes of cancer; however, a major bottleneck is the lack of efficient and standardized techniques for the...

Pharmacophore-based Identification of Potential Mutant Isocitrate Dehydrogenases I/2 Inhibitors: An Explorative Avenue in Cancer Drug Design
CONCLUSION: We investigated that ZINC9449923 of isocitrate dehydrogenase enzyme 1 and ZINC 93978407 of isocitrate dehydrogenase enzyme 2 could serve as promising candidates for the treatment of lower-grade glioma as they cross the blood-brain barrier, and present with lower toxicity levels relative to AG-881.PMID:36453510 | DOI:10.2174/1871520623666221129163001

The Combination of Zinc and Melatonin Enhanced Neuroprotection and Attenuated Neuropathy in Oxaliplatin-Induced Neurotoxicity
CONCLUSION: The combination of zinc with melatonin for the prevention approach was effective in attenuating neurotoxicity induced by oxaliplatin. The proposed mechanisms are boosting the antioxidant system and attenuating the expression of p53, GFAP, and TNF-α.PMID:36217449 | PMC:PMC9547652 | DOI:10.2147/DDDT.S385914

Aluminum oxide and zinc oxide induced nanotoxicity in rat brain, heart, and lung
This study aimed to investigate the neurotoxicity, cardiotoxicity, and lung toxicity induced by Al2O3-nps and ZnO-nps or in combination via studying changes in gene expression, alteration in cytokine production, tumor suppressor protein p53, neurotransmitters, oxidative stress, and the histological and morphological changes. Obtained results showed that Al2O3-nps, ZnO-nps and their combination cause an increase in 8-hydroxy-2´-deoxyguanosine (8-OHdG), cytokines, p53, oxidative stress, creatine kinase, norepinephrine, acetylcholine (ACh), and lipid profile. Moreover, significant changes in the gene expression of mitochondr...

Analysis of ROQUIN, Tristetraprolin (TTP), and BDNF/miR-16/TTP regulatory axis in late onset Alzheimer's disease
This study could provide a new perspective on the molecular regulatory processes associated with AD pathogenic mechanisms linked to the BDNF growth factor, although further research is needed on the possible roles of these factors in AD.PMID:36016853 | PMC:PMC9397504 | DOI:10.3389/fnagi.2022.933019

Effects of subacute cadmium exposure and subsequent deferiprone treatment on cadmium accumulation and on the homeostasis of essential elements in the mouse brain
CONCLUSION: The results demonstrated that oral administration of DFP at low doses has a better therapeutic potential for the treatment of Cd-induced brain damage compared to high doses.PMID:35985070 | DOI:10.1016/j.jtemb.2022.127062

ZNF300 enhances temozolomide resistance in gliomas by regulating lncRNA SNHG12
This study is designed to appraise the effects of ZNF300 on TMZ-resistance in gliomas. The TMZ-resistant glioma cell lines were established in U251 and A-172 cells. ZNF300 was significantly upregulated in TMZ-resistant glioma cells. ZNF300 depletion remodel TMZ sensitivity of glioma cells through inhibiting cell proliferation and promoting cell apoptosis. Meanwhile, ZNF300 depletion brought down-regulation of c-myc and SNHG12 which could be counteracted by overexpression of c-myc. Moreover, SNHG12 performed as the downstream of ZNF300 and c-myc, and overexpression of SNHG12 also could neutralize the effects of ZNF300 deple...
