Pursuit or discontinuation of anti-PD1 after 2 years of treatment in long-term responder patients with non-small cell lung cancer
CONCLUSION: The decision to continue ICI or not after 2 years of treatment depends mainly on the care center and does not seem to impact survival. Larger, randomized data sets are required to confirm this result.PMID:37720494 | PMC:PMC10501064 | DOI:10.1177/17588359231195600

Machine-Learning-Aided Prediction of Brain Metastases Development in Non-Small-Cell Lung Cancers
CONCLUSION: Analysis of the importance of input features suggests that the models are learning the relevant relationships between clinical features/development of BM. A higher number of samples is to be prioritized to improve performance. Employed prospectively at initial diagnosis, such models can help select high-risk subgroups for surveillance brain MRI.PMID:37689579 | DOI:10.1016/j.cllc.2023.08.002

Exploring beamline momentum acceptance for tracking respiratory variability in lung cancer proton therapy: a simulation study
Phys Med Biol. 2023 Aug 31. doi: 10.1088/1361-6560/acf5c4. Online ahead of print.ABSTRACTObjective
Investigating the aspects of proton beam delivery to track organ motion with pencil beam scanning therapy. Considering current systems as a reference, specify requirements for next-generation units aiming at real-time image-guided treatments.
Approach
Proton treatments for six non-small cell lung cancer (NSCLC) patients were simulated using repeated 4DCTs to model respiratory motion variability. Energy corrections required for this treatment site were evaluated for different approaches to tumour track...

Preoperative small airway dysfunction is associated with skeletal muscle loss in early-stage non-small cell lung cancer
CONCLUSION: Preoperative SAD is significantly associated with postoperative complications and SM loss in early NSCLC patients. Our results suggest that preoperative assessment of SAD may be useful for risk stratification of surgical candidates with potential for targeted interventions.PMID:37634307 | DOI:10.1016/j.clnu.2023.08.002

Abnormal Brain Glucose Metabolism Patterns in Patients with Advanced Non-Small-Cell Lung Cancer after Chemotherapy :A Retrospective PET Study
CONCLUSION: Our study revealed that patients with advanced NSCLC who underwent chemotherapy exhibited persistent abnormal brain metabolism patterns during continuous chemotherapy and these abnormalities tended to recover after completion of chemotherapy over time, but without correlation to an increasing number of chemotherapy cycles. 18F-FDG PET/CT may serve as a possible modality for evaluating brain function and guiding appropriate treatment timing.PMID:37625525 | DOI:10.1016/j.brainresbull.2023.110751

Dual-Centre Harmonised Multimodal Positron Emission Tomography/Computed Tomography Image Radiomic Features and Machine Learning Algorithms for Non-small Cell Lung Cancer Histopathological Subtype Phenotype Decoding
CONCLUSION: This study confirmed the potential of radiomic analysis on PET, CT and hybrid images for histopathological classification of NSCLC subtypes.PMID:37599160 | DOI:10.1016/j.clon.2023.08.003

First-line cemiplimab monotherapy and continued cemiplimab beyond progression plus chemotherapy for advanced non-small-cell lung cancer with PD-L1 50% or more (EMPOWER-Lung 1): 35-month follow-up from a mutlicentre, open-label, randomised, phase 3 trial
Lancet Oncol. 2023 Aug 14:S1470-2045(23)00329-7. doi: 10.1016/S1470-2045(23)00329-7. Online ahead of print.ABSTRACTBACKGROUND: Cemiplimab provided significant survival benefit to patients with advanced non-small-cell lung cancer with PD-L1 tumour expression of at least 50% and no actionable biomarkers at 1-year follow-up. In this exploratory analysis, we provide outcomes after 35 months' follow-up and the effect of adding chemotherapy to cemiplimab at the time of disease progression.METHODS: EMPOWER-Lung 1 was a multicentre, open-label, randomised, phase 3 trial. We enrolled patients (aged ≥18 years) with histologically ...

The mtDNA fragments within exosomes might be novel diagnostic biomarkers of non-small cell lung cancer
CONCLUSIONS: Changed contents of plasma and plasma exosomal mtDNAs show great potential to diagnose NSCLC, and exosomal mtDNAs might be promising biomarkers for more aggressive NSCLC.PMID:37544132 | DOI:10.1016/j.prp.2023.154718
