Hemorrhage induced by antithrombotic agents: new insights from a real-world pharmacovigilance study
CONCLUSION: We found that the risk of bleeding in patients taking Crude low molecular weight heparins was significantly higher compared to other antithrombotic agents, but with a small magnificence, which may be attributed to the severely irrational use of this medication under improper management. Statistics in days, results showed that the risk of bleeding decreased over time for warfarin and clopidogrel and increased for alteplase, nadroparin, and dipyridamole.PMID:38497691 | DOI:10.1080/14740338.2024.2327502 (Source: Expert Opinion on Drug Safety)

Contribution of platelets to disruption of the blood-brain barrier during arterial baroreflex dysfunction
CONCLUSION: During ABR dysfunction, platelets may disrupt the blood-brain barrier when CD40L on their surface activates NF-kB signaling within cerebral microvascular endothelial cells, leading to upregulation of matrix metalloprotease-9. Our findings imply that targeting CD40L may be effective against cerebral diseases involving ABR dysfunction.PMID:38493885 | DOI:10.1016/j.mvr.2024.104681 (Source: Cell Research)

Relationship between in-stent restenosis following carotid artery stenting and platelet reactivity to clopidogrel
CONCLUSIONS: High PRU (≥220) and low IR (≤14.5%) are related to ISR R3 following CAS, which may cause ipsilateral ischemic stroke.PMID:38490734 | DOI:10.1136/jnis-2024-021445 (Source: Atherosclerosis)

Viruses, Vol. 16, Pages 450: Small Molecules for the Treatment of Long-COVID-Related Vascular Damage and Abnormal Blood Clotting: A Patent-Based Appraisal
iano Altomare People affected by COVID-19 are exposed to, among others, abnormal clotting and endothelial dysfunction, which may result in deep vein thrombosis, cerebrovascular disorders, and ischemic and non-ischemic heart diseases, to mention a few. Treatments for COVID-19 include antiplatelet (e.g., aspirin, clopidogrel) and anticoagulant agents, but their impact on morbidity and mortality has not been proven. In addition, due to viremia-associated interconnected prothrombotic and proinflammatory events, anti-inflammatory drugs have also been investigated for their ability to mitigate against immune dysregulation du...

Development of a routine bedside CYP2C19 genotype assessment program  for antiplatelet therapy guidance in a community hospital catheterization laboratory
Abstract Genotype based personalized antiplatelet therapy in the setting of percutaneous coronary intervention (PCI) has been studied in clinical trials. Despite the demonstrated risk associated withCYP2C19 loss-of-function(LoF) carriage in clopidogrel-treated PCI patients, real-world implementation of genotyping for PCI has been low. The goal of the current study was to provideCYP2C19 genotype information to the interventionalist prior to the completion of the catheterization to facilitate immediate personalized antiplatelet therapy. Routine personalization of P2Y12 inhibitor therapy for PCI in a community hospital cardia...

Generalisability of trials on antithrombotic treatment intensification in patients with cardiovascular disease
Conclusion A large proportion of contemporary CVD patients would be eligible for intensified antithrombotic treatment trials, with mostly similar adjusted event risks to ineligible patients. Trial-based guideline recommendations are largely applicable to real-world patients. (Source: Heart)
