Improvement extraction of phenolic compound by using ion exchange chromatography and evaluation of biological activities of polyphenol-enriched fraction of Rosa canina fruits

Rosa canina has been traditionally known as a medicinal plant. Its fruits (Rose hip) have been used in several food products, as well as perfumery and cosmetics industries.The aim of this study was to prepare an enriched polyphenolic fraction from R. canina in addition to its biological activities.Poly phenolic enriched fraction was prepared using Amberlite XAD-7 for removing unwanted components. Phenols, flavonoids and anthocyanins content analyses showed that they were increased significantly compared to the extract. HPLC analysis showed that this fraction is rich source of ascorbic acid. The results of DPPH, FRAP, ABTS and nitric oxide assay confirmed that the antioxidant activities of the fraction had been increased in comparison with the extract. The ORAC assay and cellular antioxidant activity of the fraction confirmed its potential antioxidant activity as well. This fraction showed xanthine oxidase inhibitory activity at 100µg/ml concentration. Comet assay analysis revealed that this fraction at 25-100µg/ml concentrations protects human lymphocytes against H2O2-induced DNA damages.Taken together, this study suggests that fruit of Rosa canina could be considered as potential antioxidant, xanthin oxidase inhibitor and a antigenotoxic source, and application of Amberlite XAD-7 improves extraction efficiencies by means of enrichment of phenolic compounds in this plant.
