CLL-390 Persistence and Time to Next Treatment With Ibrutinib Treatment in Patients With Chronic Lymphocytic Leukemia/Small Lymphocytic Lymphoma Including Patients at High Risk for Atrial Fibrillation or Stroke: A Real-World Study
Context: Atrial fibrillation (AF) is a recognized adverse consequence associated with all Bruton's tyrosine kinase inhibitors used to treat chronic lymphocytic leukemia (CLL)/small lymphocytic lymphoma (SLL); however, real-world time to discontinuation (TTD) and time to next treatment (TTNT) of CLL/SLL patients with a high baseline AF risk remain unknown. Objective: To describe TTNT and TTD for CLL/SLL patients who initiated a first-line (1L) or second-or-later-line (2L+) therapy with ibrutinib or other regimens, in all patients and for subsets of patients with high baseline risk of AF/AF-related events.

Severe, Fatal Methotrexate-related Neurotoxicity in 2 Adolescent Patients With ALL
Neurotoxicity is a well-documented adverse effect of methotrexate in the treatment of pediatric cancers. The spectrum of symptoms is broad, can include stroke-like episodes and seizures, and classically resolves within days. The majority of patients tolerate subsequent doses without recurrence of symptoms. Risk of thromboembolic and bleeding outcomes following hematological cancers: a Danish population-based cohort study.
CONCLUSIONS: Approximately 2 out of 10 hematological cancer patients experienced MI, ischemic stroke, VTE, or bleeding requiring hospital contact within 10 years. The hematological cancer cohort had higher hazards of MI, ischemic stroke, VTE, and bleeding requiring hospital contact than a general population comparison cohort. A Retrospective Study of Comorbidities and Complications in Elderly Acute Myeloid Leukemia Patients in the United States
There is limited information on the comorbidities and complications in elderly AML patients. In a sample of 3,911 AML patients ( ≥ 65 years) identified from the SEER-Medicare database, AML patients had more comorbidities, higher rates of complications, and a higher risk of developing cardiovascular disease, type 2 diabetes mellitus, and stroke compared to matched non-cancer controls.

Outcome of 472 Chronic Myeloid Leukemia Patients Treated with Frontline Nilotinib: A Gimema CML WP Analysis
CONCLUSION: Nilotinib as first-line treatment of newly diagnosed CP CML pts showed high rates of deep molecular responses, few progressions to AP/BP, and a high OS. Deep molecular response were similar in all age groups; as expected, ATEs were more frequent in pts > 60 y. Increased Risk of Lymphoid Malignancy in Patients with Herpes Zoster: A Longitudinal Follow-up Study Using a National Cohort Study

Medical Conditions Among Survivors of Adolescent and Young Adult Non-Hodgkin Lymphoma (NHL), Acute Lymphoblastic Leukemia (ALL) and Acute Myeloid Leukemia (AML)
Conclusion: This study found that sociodemographic factors were associated with the risk of developing medical conditions in AYA NHL, ALL and AML survivors. As expected, the risk of medical conditions varied by cancer type and treatment, with those undergoing SCT having a higher risk of medical conditions regardless of cancer type. NHL and ALL survivors who were uninsured or publicly insured were at a consistently higher risk of developing medical conditions, as were Hispanic ALL survivors and Black AML survivors.
