Applications of tissue-specific and cancer-selective gene promoters for cancer diagnosis and therapy
Adv Cancer Res. 2023;160:253-315. doi: 10.1016/bs.acr.2023.03.005. Epub 2023 Apr 20.ABSTRACTCurrent treatment of solid tumors with standard of care chemotherapies, radiation therapy and/or immunotherapies are often limited by severe adverse toxic effects, resulting in a narrow therapeutic index. Cancer gene therapy represents a targeted approach that in principle could significantly reduce undesirable side effects in normal tissues while significantly inhibiting tumor growth and progression. To be effective, this strategy requires a clear understanding of the molecular biology of cancer development and evolution and develo...

Long-term effects of a fat-directed FGF21 gene therapy in aged female mice
Gene Ther. 2023 Sep 12. doi: 10.1038/s41434-023-00422-0. Online ahead of print.ABSTRACTFibroblast growth factor 21 (FGF21) has been developed as a potential therapeutic agent for metabolic syndromes. Moreover, FGF21 is considered a pro-longevity hormone because transgenic mice overexpressing FGF21 display extended lifespan, raising the possibility of using FGF21 to promote healthy aging. We recently showed that visceral fat directed FGF21 gene therapy improves metabolic and immune health in insulin resistant BTBR mice. Here, we used a fat directed rAAV-FGF21 vector in 17-month-old female mice to investigate whether long-te...

Apolipoprotein A-I vascular gene therapy reduces vein-graft atherosclerosis
Mol Ther Methods Clin Dev. 2023 Aug 22;30:558-572. doi: 10.1016/j.omtm.2023.08.018. eCollection 2023 Sep 14.ABSTRACTCoronary artery venous bypass grafts typically fail because of atherosclerosis driven by lipid and macrophage accumulation. Therapy for vein-graft atherosclerosis is limited to statin drugs, which are only modestly effective. We hypothesized that transduction of vein-graft endothelium of fat-fed rabbits with a helper-dependent adenovirus expressing apolipoprotein AI (HDAdApoAI) would reduce lipid and macrophage accumulation. Fat-fed rabbits received bilateral external jugular vein-to-carotid artery interposit...

Up-to-date molecular medicine strategies for management of ocular surface neovascularization
Adv Drug Deliv Rev. 2023 Sep 7:115084. doi: 10.1016/j.addr.2023.115084. Online ahead of print.ABSTRACTOcular surface neovascularization and its resulting pathological changes significantly alter corneal refraction and obstruct light path to the retina, and hence is a major cause of vision loss. Various factors such as infection, irritation, trauma, dry eye, and ocular surface surgery trigger neovascularization via angiogenesis and lymphangiogenesis dependent on VEGF-related and alternative mechanisms. Recent advances in antiangiogenic drugs, nanotechnology, gene therapy, surgery equipment and techniques, animal models, and...

Stimuli-Responsive Hydrogel Microcapsules Harnessing the COVID-19 Immune Response for Cancer Therapeutics
We present the development of stimuli-responsive microcapsules, loaded with a viral immunogenetic agent, harnessing the immune response against the Coronavirus Disease 2019, COVID-19, to selectively attack liver cancer cells (hepatoma) or recognize breast cancer or hepatoma, by expression of green fluorescence protein, GFP. The pH-responsive microcapsules, modified with DNA-tetrahedra nanostructures, increased hepatoma permeation by 50%. Incorporation of a GFP-encoding lentivirus vector inside the tumor-targeting pH-stimulated miRNA-triggered and Alpha-fetoprotein-dictated microcapsules enables the demonstration of neoplas...

Cold atmospheric plasma mediated cell membrane permeation and gene delivery-empirical interventions and pertinence
Adv Colloid Interface Sci. 2023 Aug 28;320:102989. doi: 10.1016/j.cis.2023.102989. Online ahead of print.ABSTRACTDelivery of genetic material to cells is an integral tool to analyze and reveal the genetic interventions in normal cellular processes and differentiation, disease development and for gene therapy. It has profound applications in pharmaceutical, agricultural, environmental and biotechnological sectors. The major methods relied for gene delivery or transfection requires either viral vectors or xenogenic carrier molecules, which renders probabilistic carcinogenic, immunogenic and toxic effects. A newly evolved phy...
