Expanding Biology of PCSK9: Roles in Atherosclerosis and Beyond
AbstractPurpose of ReviewSince the discovery of PCSK9 in 2003, this proprotein convertase was shown to target specific receptors for degradation in endosomes/lysosomes, including LDLR and other family members and hence to enhance the levels of circulating LDL-cholesterol (LDLc). Accordingly, inhibitors of PCSK9, including monoclonal antibodies blocking its circulating activity and siRNA silencers of its hepatic expression, are now used in clinics worldwide to treat hypercholesterolemia patients effectively and safely in combination with statins and/or ezetimibe. These powerful treatments reduce the incidence of atheroscler...

The performance and perspectives of dendritic cell vaccines modified by immune checkpoint inhibitors or stimulants
Biochim Biophys Acta Rev Cancer. 2022 Jul 21:188763. doi: 10.1016/j.bbcan.2022.188763. Online ahead of print.ABSTRACTTherapeutic dendritic cell (DC) vaccines stimulate the elimination of tumor cells by the immune system. However, while antigen-specific T cell responses induced by DC vaccines are commonly observed, the clinical response rate is relatively poor, necessitating vaccine optimization. There is evidence that the suppression of DC function by immune checkpoints hinders the anti-tumor immune responses mediated by DC vaccines, ultimately leading to the immune escape of the tumor cells. The use of immune checkpoint i...

Abstracts of Presentations at the Association of Clinical Scientists 143 < sup > rd < /sup > Meeting Louisville, KY May 11-14,2022
Conclusion: These assays are suitable for routine diagnostic. The UltraFast NextGenPCR is the fastest with average time (30mins), followed by Agilent (2 hrs) and MassArray (6hrs). Upon completion of this activity, participants should be able to examine, measure and compare results from different assays for SARS detection, evaluate and diagnose accurately, as well as being able to plan, organize and recommend a diagnostic procedure for diagnostic laboratory. Key words: SARS-CoV-2, RNA extraction, RT-PCR, limit of detection, quantification cycle, COVID-19, in vitro diagnostic tests, Agilent, Massarray, Ultrafast. [20] From t...

Dual Blockade of PD-1 and LAG3 Immune Checkpoints Increases Dendritic Cell Vaccine Mediated T Cell Responses in Breast Cancer Model
CONCLUSION: These findings strongly suggest that TMC-DS-L NPs loaded with siRNA could act as a novel tool in inhibiting the expression of immune checkpoints in the tumor microenvironment. Also, combination therapy based on inhibition of PD-1 and LAG3 in combination with DC vaccine is an effective method in treating cancer that needs to be further studied.PMID:35715669 | DOI:10.1007/s11095-022-03297-9
