Current advancements in self-assembling nanocarriers-based siRNA delivery for cancer therapy
Colloids Surf B Biointerfaces. 2022 Nov 2;221:113002. doi: 10.1016/j.colsurfb.2022.113002. Online ahead of print.ABSTRACTDifferent therapeutic practices for treating cancers have significantly evolved to compensate and/or overcome the failures in conventional methodologies. The demonstrated potentiality in completely inhibiting the tumors and in preventing cancer relapse has made nucleic acids therapy (NAT)/gene therapy as an attractive practice. This has been made possible because NAT-based cancer treatments are highly focused on the fundamental mechanisms - i.e., silencing the expression of oncogenic genes responsible fo...

Recent advances in siRNA delivery systems for prostate cancer therapy
Curr Pharm Biotechnol. 2021 Jun 15. doi: 10.2174/1389201022666210615123211. Online ahead of print.ABSTRACTThe critical problems of conventional prostate cancer therapeutic strategies like nonspecific toxicity and multi-drug resistance prompted the development and application of countless nanoparticle-based siRNA therapeutics. The main challenges to siRNA-based therapeutics becoming a new paradigm in the treatment of prostate cancer stem from the lack of safe and effective delivery systems, immune system stimulation, poor knowledge of nano-bio interactions, and limitations concerning designing, manufacturing, clinical trans...

Targeting clusterin induces apoptosis, reduces growth ability, invasion and mediates sensitivity to chemotherapy in human cancer in human osteosarcoma cells.
CONCLUSIONS: Overexpression of sCLU was significantly correlated with metastasis and chemosensitivity in osteosarcoma cells. The described sCLU-specific siRNA that can potently silence sCLU gene expression may thus prove valuable agents during antitumor therapy. PMID: 31433751 [PubMed - as supplied by publisher]

Toxicological exploration of peptide-based cationic liposomes in siRNA delivery.
Abstract The toxicology of cationic liposomes was explored to advance clinical trials of liposome-mediated gene therapy through the analysis of a peptide cationic liposome with DOTAP as a positive control. We first investigated the delivery of luciferase siRNA by several peptide liposomes in mice bearing lung cancer A549 cell xenografts. Of these, a cationic liposome (CDO14) was selected for further investigation. CDO14 efficiently mediated IGF-1R-siRNA delivery and inhibited the growth of the A549 cell xenografts. The in vivo toxicity and toxicological mechanisms of the selected liposome were evaluated to assess ...
