A Transcriptional Regulation Bioinformatics Pipeline to Predict Co-Regulated Genes in Vascular Smooth Muscle Cell Phenotypic Transitions During Atherosclerosis
This study proposes a novel systems biology pipeline that integrates experimental and computational tools with large datasets measuring the transcriptomes and epigenomes of phenotypically transitioning VSMCs. Our pipeline allows users to identify candidate transcription factors that co-regulate mouse or human gene sets controlling specific VSMC phenotypic transitions during atherosclerosis. The computational algorithms developed in this workflow integrate an array of publicly available databases, such as the Ensembl genome database and the JASPAR transcription factor database. To demonstrate the translational potential of ...

Cannabinoid receptor 1 antagonist genistein attenuates marijuana-induced vascular inflammation
Cell. 2022 Apr 29:S0092-8674(22)00443-3. doi: 10.1016/j.cell.2022.04.005. Online ahead of print.ABSTRACTEpidemiological studies reveal that marijuana increases the risk of cardiovascular disease (CVD); however, little is known about the mechanism. Δ9-tetrahydrocannabinol (Δ9-THC), the psychoactive component of marijuana, binds to cannabinoid receptor 1 (CB1/CNR1) in the vasculature and is implicated in CVD. A UK Biobank analysis found that cannabis was an risk factor for CVD. We found that marijuana smoking activated inflammatory cytokines implicated in CVD. In silico virtual screening identified genistein, a soybean iso...

Therapeutic Potential of Stem Cell-Derived Extracellular Vesicles on Atherosclerosis-Induced Vascular Dysfunction and Its Key Molecular Players
Front Cell Dev Biol. 2022 Mar 18;10:817180. doi: 10.3389/fcell.2022.817180. eCollection 2022.ABSTRACTAtherosclerosis is a progressive, chronic inflammatory disease of the large arteries caused by the constant accumulation of cholesterol, followed by endothelial dysfunction and vascular inflammation. We hypothesized that delivery of extracellular vesicles (EVs), recognized for their potential as therapeutic targets and tools, could restore vascular function in atherosclerosis. We explored by comparison the potential beneficial effects of EVs from subcutaneous adipose tissue stem cells (EVs (ADSCs)) or bone marrow mesenchyma...

Novel Mechanisms of Exosome-Mediated Phagocytosis of Dead Cells in Injured Heart
Conclusions: Our data demonstrates that exosome-associated MFGE8 on one hand enhances opsonization of dead cells and on the other activates phagocytic signaling thus augmenting removal of apoptotic cells, resolution of inflammation and therefore efficient cardiac recovery after injury.PMID:34623174 | DOI:10.1161/CIRCRESAHA.120.317900

The Unfolded Protein Response as a Compensatory Mechanism and Potential Therapeutic Target in PLN R14del Cardiomyopathy
Conclusions: Together, these findings suggest that the UPR exerts a protective effect in the setting of PLN R14del cardiomyopathy and that modulation of the UPR might be exploited therapeutically.PMID:33928785 | DOI:10.1161/CIRCULATIONAHA.120.049844

Integrative Genomic Analysis Reveals Four Protein Biomarkers for Platelet Traits.
Conclusions: We identified four proteins that are causally linked to platelet counts. These proteins may also have roles in the pathogenesis of CVD via a platelet/blood coagulation-based mechanism. PMID: 32781905 [PubMed - as supplied by publisher]

From traditional pharmacological towards nucleic acid-based therapies for cardiovascular diseases
AbstractNucleic acid-based therapeutics are currently developed at large scale for prevention and management of cardiovascular diseases (CVDs), since: (i) genetic studies have highlighted novel therapeutic targets suggested to be causal for CVD; (ii) there is a substantial recent progress in delivery, efficacy, and safety of nucleic acid-based therapies; (iii) they enable effective modulation of therapeutic targets that cannot be sufficiently or optimally addressed using traditional small molecule drugs or antibodies. Nucleic acid-based therapeutics include (i) RNA-targeted therapeutics for gene silencing; (ii) microRNA-mo...
