An investigation of excipients for a stable Orf viral vector formulation
In this study, the non-supplemented ORFV itself showed a very high stability at storage temperatures up to 28°C, with a linear titer loss of 0.10 log infectious particles per day at 4°C over a period of five weeks. To prolong this inherent stability, thirty additives, i.e., detergents, sugars, proteins, salts, and buffers as well as amino acids, were tested for their time- and temperature-dependent influence on the ORFV infectivity. A stabilizing effect on the infectivity was identified for the addition of all tested proteins, i.e., gelatine, bovine serum albumin, and recombinant human serum albumin (rHSA), of several su...

Physical Characteristics and Stability Profile of Recombinant Plasmid DNA within a Film Matrix
The objective of these studies was to identify a film-based formulation that preserved transfection efficiency of plasmids at 25°C. A model plasmid, pAAVlacZ, was used for these studies. Transfection efficiency and agarose gel electrophoresis were utilized to assess bioactivity and changes in physical conformation of plasmid during storage. An amino acid, capable of sustaining a positive charge while supporting an alkaline environment within the film matrix, preserved transfection efficiency for 9 months at 25°C. Addition of sugar and a plasticizer to the formulation preserved the plasmid in an amorphous state and improv...

Lignocellulosic biomass-based glycoconjugates for diverse biotechnological applications
Biotechnol Adv. 2023 Jul 17:108209. doi: 10.1016/j.biotechadv.2023.108209. Online ahead of print.ABSTRACTGlycoconjugates are the ubiquitous components of mammalian cells, mainly synthesized by covalent bonds of carbohydrates to other biomolecules such as proteins and lipids, with a wide range of potential applications in novel vaccines, therapeutic peptides and antibodies (Ab). Considering the emerging developments in glycoscience, renewable production of glycoconjugates is of importance and lignocellulosic biomass (LCB) is a potential source of carbohydrates to produce synthetic glycoconjugates in a sustainable pathway. I...

Evaluation of structural modification induced activation of pneumococcal polysaccharide by GC-MS for the conjugate vaccine
Carbohydr Res. 2023 Jun 25;531:108878. doi: 10.1016/j.carres.2023.108878. Online ahead of print.ABSTRACTPolysaccharide (Ps) activation evaluation is an imperative quality attribute in a conjugate vaccine. Pneumococcal polysaccharide (PnPs) serotypes 5, 6B, 14, 19A and 23F were cyanylated for 3 and 8 min. The cyanylated and non-cyanylated polysaccharides were methanolysed and derivatized to assess the activation of each sugar by GC-MS. The activation of 22 and 27% serotype 6B and 11 and 36% in serotype 23 F Ps at 3 and 8 min respectively showed controlled conjugation kinetics with CRM197 carrier protein estimated by SEC-HPL...

Fight Aging! Newsletter, June 19th 2023
In conclusion, among Swedish middle-aged subjects, nearly two-thirds showed complete fatty degeneration of thymus on CT. Age-Related Dysfunction of Water Homeostasis https://www.fightaging.org/archives/2023/06/age-related-dysfunction-of-water-homeostasis/ Dehydration can be an issue in older people. As in every complex system in the body, the mechanisms by which hydration is regulated become dysfunctional with advancing age. Researchers here look at the brain region responsible for regulating some of the response to dehydration, cataloging altered gene expression in search of the more important mechani...

Stereoselective Synthesis of the O-antigen of A. baumannii ATCC 17961 Using Long-Range Levulinoyl Group Participation
Angew Chem Int Ed Engl. 2023 Jun 16:e202306971. doi: 10.1002/anie.202306971. Online ahead of print.ABSTRACTHerein, we described the first synthesis of pentasaccharide and decasaccharide of A. baumannii ATCC 17961 O-antigen for developing a synthetic carbohydrate-based vaccine against A. baumannii infection. The efficient synthesis of the rare sugar 2,3-diacetamido-glucuronate was achieved using our recently introduced organocatalytic glycosylation method. We found, for the first time, that long-range levulinoyl group participation via a hydrogen bond can result in a significantly improved β-selectivity in glycosylations. ...

Synthesis of immunostimulatory saponins: A sweet challenge for carbohydrate chemists
Carbohydr Res. 2023 May 23;530:108851. doi: 10.1016/j.carres.2023.108851. Online ahead of print.ABSTRACTSaponins are a large family of natural glycosides showing a wide range of biological activities. Current research efforts on saponins as vaccine adjuvants have been mainly focused on the development of synthetic analogs. By mimicking the immunomodulatory saponins from Quillaja saponaria (QS), less complex and readily accessible analogs have been synthesized to improve the industrial applicability and efficacy of saponins as vaccine adjuvants. Through the exploration of several structural modifications on the skeleton of ...
